- Description
- The Perinatology Research Branch (PRB) of the National Institute of Child Health and Human Development (NICHD) is planning to contract with Lark Technologies, Inc., Houston, Texas for studies using Real-Time Quantitative Polymerase Chain Reaction (PCR) to quantify the amount of specific mRNA to confirm variations in expression levels of candidate genes identified in microarray expression profiles. These genes may be involved in the physiological and pathological processes of pregnancies. The contractor must have the capability to receive total RNA (isolated and purified by the PRB), reverse transcribe it to cDNA, have currently available a large menu of optimized QPCR assays for human cytokines, design new QPCR assays for the genes of interest to the PRB and carry out the real-time QPCR for these genes and for housekeeping genes or 18 S ribosomal RNA as controls in triplicate assays, and report the relative, standardized levels of expression for each gene. The contractor shall provide the following services: 1) Provide optimized QPCR assays for quantification of mRNA levels of human cytokines. 2) Generate cDNA from RNA samples using reverse transcriptase reaction. 3) Perform real-time QPCRs and use 18 S ribosomal RNA as an internal standard for each RNA sample. 4) Design new real-time QPCR assays for genes of interest to the PRB and not yet part of the contractor's product line. 5) Perform real-time QPCRs with new assays and use 18 S ribosomal RNA as an internal standard for each RNA sample. 6) Perform quality control with positive and negative control elements placed on each QPCR plate. and 7) Report results along with details on the assays (including primer and probe sequences as well as QPCR conditions) in the form of Microsoft Excel to PRB appointees. The purpose of this synopsis is to determine whether there are other sources with the requisite qualifications to perform the proposed study. Any organization capable of performing the work should submit a statement of capability with documentation supporting its capabilities within 45 days from the date of publication of this announcement.
