The Perinatology Research Branch (PRB) of the National Institute of Child Health and Human Development (NICHD) has a need for Microarray Services. In accordance with 41 U.S.C. 253 (c) 1 as set forth in FAR 6.302-1(a)(2)(ii), the NICHD has determined that Incyte Genomics, Inc., Palo Alto, California, is the only source capable of using microarray technology to screen characterized cDNA sequences to detect variations in expression levels of candidate genes which may be involved in the physiological and pathological processes of pregnancies. In addition, they also have the capability to receive total RNA (isolated and purified by the PRB), isolate polyA+RNA from the total RNA, reverse transcribe it to cDNA, construct high-density microarrays consisting of cDNA clones and report the identity of the genes and magnitude of change in expression in a pair-wise comparison. Any source who believes that they have the above capabilities should demonstrate they can perform the following: 1) Prepare high-density microarrays on glass slides with a minimum of 9,000 sequence-verified cDNA clones from the UniGene collection. 2) Isolate polyA+RNA from total RNA samples provided by RFB. 3) Generate probes from RNA samples using reverse transcriptase reaction. 4) Hybridize cDNA probes generated in step 3 to microarrays in a pair-wise manner. 5) Perform washes of the hybridized microarrays. 6) Conduct image analyses and format data for GEMTools software analyses. and 7) Perform quality control with control elements placed on each microarray. The purpose of this synopsis is to determine whether there are other sources that can perform the work described above.
