Contracting Officer The National Cancer Institute (NCI) plans
to procure a Symphony Multiplex Peptide Synthesizer,computer, color
monitor, printer, 12 glass reaction vessels, solvent cabinent, waste
tanks, a start-up kit, a 20L vessel assembly and on site installation
and training from Rainin Instrument Company, P.O. Box 4026, Mack Road,
Woburn, MA 01801. A multiplex peptide synthesizer is needed to perform
several structural studies in one of the NCI laboratories. Since there
is a heavy volume of syntheses across different reaction times and
protocols, a system that allows for simultaneous production of
completely independent compounds and in different quantities is
required. This will enable work to be completed in a more timely,
flexible, safe and efficient manner. The required system shall perform
rapid and efficient solid phase synthesis of large numbers of
compounds with different sequencers. The instrument shall allow at
least 12 reaction vessels to produce compounds quickly and in ample
numbers with many reaction vessels. In addition, the valve system shall
be a patented multiplexing matrix style that permits different reagents
to be transferred to different reaction vessels (Rvs) without
cross-contamination. The instrument shall allow different reaction
times and protocols to be applied for each synthesis, allowing the
instrument to be used for simultaneous production of completely
independent compounds. It shall enable compounds of differing lengths
to be synthesized simultaneously in different quantities. The reagents
shall be dispensed from common reservoirs for the synthesis of all
compounds. The system shall also allow for the use of bulk reagents to
be used to prepare solutions for 20 or more reagent reservoirs. The
instrument shall also enable the reaction vessels to be agitated during
coupling to allow the synthesis of larger compounds and permit the
solution to be only moderately concentrated for easy dissolution of
reagents. The machine shall also provide automated cleavage to deliver
the product from each synthesis directly to a centrifuge tube in a
vented compartment in order to be set up for further work up of the
material. A variable scale of 0.005 mM to 0.35 mM or greater shall be
provided with the equipment to enable the synthesis of both large and
small quantities. All vessels and steps for different peptides being
processed shall be acted upon simultaneously, as opposed to
independently. In addition, if peptides are done at varying times,
there shall be no waiting or stoppage of the synthesis to remove
completed reaction vessels and to replace them with a blank. In
addition, to improve product quality, the couplings shall occur under
an inert atmosphere of Nitrogen. In summary, the instrument shall
possess the following features: 1) At least 12 reaction vessels; 2) The
capability to perform 12 different coupling protocols simultaneously;
3) Liquid sensors; 4) The capability of simultaneous solution transfer
via a matrix valve; 5) The ability to perform cleavage via a fully
automated and enclosed cleavage system built into the instrument; 6)
The capability to measure delivered volumes of reagents via a sample
loop; 7) The ability to accomplish syntheses completely independent of
one another but simultaneously; 8) Agitation completed through
nitrogen; 9) The ability to transfer solutions by nitrogen; 10) The
ability to use standard protocols or customized reaction protocols for
each individual synthesis; 11) The ability to use a conventional
synthesizer; and 12) The capability of being used/compatible with
standard laboratory ventilation systems. Rainin Instrument is the only
source known to the NCI Researcher that can meet most of the above
specifications. This notice of intent is not a request for competitive
quotation. However, if any interested party believes it can meet the
above requirements, it may submit a statement of capabilities. The
capability statement and any other furnished information must be in
writing and must contain material in sufficient detail to allow NCI to
determine if the party can fully meet the requirements. Capability
statements must be received in the contracting office by 3:00 p.m., EST

responses to this notice is solely within the discretion of the
