as Operations and Technical Support contractor to the National Cancer
Institute (NCI) for the Frederick Cancer Research and Development
Center is seeking a subcontractor which, following protocols provided
by the National Cancer Institute (NCI), will generate supernatants
containing patient specific immunoglobulins (PSI) for use in the
production of experimental vaccines for B-cell lymphoma. These
supernatants containing PSI will come from hybridoma cell lines
generated by the fusion of the patients tumor cells with a mouse-human
heterohybridoma cell line. Supernatants from these fusion lines will
contain the same PSI as the original tumor cells. The subcontractor
will be required to: 1) expand the fusion cell lines, 2) generate
supernatants containing PSI, and 3) assay these supernatants for PSI,
all under well controlled conditions using either Good Laboratory
Practices (GLP) or Good Manufacturing Practices (GMP) as necessary.
Supernatants will be generated in small hollow-fiber bioreactors
according to a protocol provided by the NCI, and should be shown by the
contractor to be sterile, mycoplasma-free, and non-reactive in an in
vitro adventitious-virus assay. It is anticipated that the
subcontractor will produce approximately 50 of these supernatants
containing PSI per year, with each supernatant containing between
100-200 mg of PSI. The subcontractor will not be responsible for the
generation of the fusion cell lines or the purification of the PSI.
