65 -- DNA SEQUENCING KITS SOL RFQ90038-NS DUE 022599 POC Patricia Haun, Purchasing Agent, 301-402-4509 The National Cancer Institute (NCI) on behalf of the Division of Intramural Research, Basic Neuroscience Program, National Institute of Neurological Disorders and Stroke (NINDS) plans to procure BigDye Terminator DNA sequencing TF kits from PE Biosystems, Applied Biosystems Div., 850 Lincoln Ctr. Dr., Foster City, CA 94404. The NINDS DNA Sequencing Facility provides DNA sequencing as a service to researchers in the NINDS Division of Intramural Research (DIR). This reaction kit is available with a single energy transfer molecule using a donor fluorescein and an acceptor dichlororhodamine coupled by a highly efficient energy transfer linker. This feature provides a very bright signal in which longer read lengths occur. The sequencing kits use one (1) tube per sample instead of the usual four (4) tubes per sample required for primer sequencing chemistry. The Ready Reaction kit format contains BidDye terminators, deoxynucleoside triphosphates, AmpliTaq FS enzyme, magnesium chloride, and reaction buffer premixed in a single tube. In addition, dichlororhodamine dyes have better spectral separation at their maximum excitation efficiency. These sequencing products show much reduced background noise and are specifically designed to enhance the results on ABI Prism Genetic Analysis systems. Also the kits used in conjunction with AmpliTaq FS enzyme results in more even peak patterns, cleaner signal and greater base calling accuracy. Two machines used by NINDS currently utilize the PE Biosystems BigDye Terminator reagents. To use different reagents on another machine would introduce unnecessary variables into the work being performed and could affect the research results. A standing order is anticipated for one (1)box per month of the TF Kit, with BigDye Terminator Ready Reaction -- 100. PE Biosystems is the only source known to the NINDS that can meet the above requirements in conjunction with the existing PE ABI 373 and PE ABI 377 systems. If any interested party believes it can provide the above requirements, itmay submit a statement of capabilities. The statement of capabilities and any other information furnished must be in writing and must contain material in sufficient detail to allow NCI to determine if the party can meet this requirement.
