The VA Medical Center in Gainesville, Florida intends to negotiate a
sole source contract with the University of Florida, Department of
Medicine beginning the date of award for one year with two optional
years for renewal. The overall objective of this study entitled,
Biophysical analysis of marine natural products that mimic the
actin-binding properties of gelsolin with the Prinicipal Investigator
being Dr. Michael Bubb is to define quantitative aspects of the
interaction between marine natural products and actin using biophysical
techniques. Time dependent fluorescence anisotropy, fluorescence energy
transfer, analytical ultracentrifugation and 3-dimensional molecular
modeling will be used to assess on-and off-rates, stoichiometry and
structural relationships. The rationale for this proposed study is to
identify lead compounds to be developed as anti-tumor agents mimicing
the function of the tumor suppressor, gelsolin. In order for this to
proceed in a rationale manner, a quantatative comparison of marine
natural products must be used to identify similarities and differences
between the activities of gelsolin and the activities of each marine
