The Food and Drug Administration [FDA], Center for Biologics and Evaluation [CBER], intends to award a purchase order for a LC32 Lightcycler from Appropriate Technical Resources, Inc. The Lightcycler shall integrate a 32 sampled micro-volume fluorimeter with an air thermal cycler to deliver accurate and high-speed temperature control. Equipment shall allow researcher to watch in real time the quantitative replication of DNA or RNA, and continuous monitoring of the thermal cycling reaction with 30 replications of DNA to be accomplished in less than 10 minutes. The Lightcycler shall perform both amplification and separate product analysis in application; the cuvettes shall double as natural cuvetters for fluorescence acquisition due to light-piping at the tip of the capillary curvettes. Equipment shall monitor the fluorescence from the double-standard DNA dye SYBR Green [Molecular Probles], coupled with a novel approach to product differentiation by analysis of product melting curves, allowing for inexpensive quanification of low initial template copy number. Probes shall be designed for gene quanification to the single copy level in less than 10 minutes thus allowing for economical pathogen and/or mutation detection. Equipment shall perform 30 cycles in less than 10 minutes. Equipment shall be as follows: temperature controlled micro-volume fluorimeter, 32 sample, real-time display of fluorescence and temperature data in Windows '95; storage shall be text file; composite glass/plastic reaction vessel loaded by centrifugation; 1-10 ul; up to three [3] color optics modules, excitation wavelengths from 450-670 nm., emission wavelengths from 500-750 nm., 60-1000 msec/sample, 6 seconds for 32 samples [or less depending on sample count]: 1] continuous [single sample], 2] once per cycle, and 3] continuous scanning mode [frequency dependent on sample count]; integrated windows '95 program for programming, running reactions, fluorescence data acquisition, and data analysis; up to 100 target temperaturesper cycle program; target temperatures consist of temperature, time, slope, step functions, and acquisition mode; thousands of named sequential programs with up to 100 cycle programs per sequence; room temperature to 120 degrees C; within sample and between shall be + [plus is underlined], 0.2 degrees C; Ramp Rate -- 0.1 -- 20 degrees C/sec, 90-130 V, 50/60 HZ, 5.8 Amp, 700 Wattt, 200-240 V, 50/60 HZ, 2.9 Amp, 700 Watt.
