- DESCRIPTION: This is a Sources Sought notice. This is NOT a solicitation for proposals, proposal abstracts, or quotations. The purpose of this notice is to obtain information regarding the availability and capability of all qualified sources to provide equipment for this requirement. This notice is issued to help determine the availability of qualified companies technically capable of meeting the Government requirement and to determine the method of acquisition.� It is not to be construed as a commitment by the Government to issue a solicitation or ultimately award a contract.� Responses will not be considered as proposals or quotes.� No award will be made as a result of this notice.� The Government will NOT be responsible for any costs incurred by the respondents to this notice.� This notice is strictly for research and information purposes only. BACKGROUND INFORMATION AND OBJECTIVE: The National Institute on Neurological Disease and Stroke (NINDS) Inherited Neurodegenerative Disease Unit (INDU) and National Institutes of Health (NIH) is seeking Capability Statements from organizations with the demonstrated capability of providing equipment to support NINDS with acquiring an Optical Pooled Screen Microscope which will be used to identify converging mechanisms of disease using sophisticated optical pooled screening approaches, in which iPSC derived neurons are manipulated with CRISPR-based techniques at the single cell level, followed by automated acquisition of relevant cellular phenotypes via microscopy, followed by in situ sequencing (ISS) of sgRNAs. This microscope will be used to identify converging mechanisms of disease using sophisticated optical pooled screening approaches, in which iPSC derived neurons are manipulated with CRISPR-based techniques at the single cell level, followed by automated acquisition of relevant cellular phenotypes via microscopy, followed by in situ sequencing (ISS) of sgRNAs. This equipment is essential for the mission of the INDU, to determine upstream causes of neurodegenerative disease and identify potential therapeutic targets, in line with the overall mission of NINDS to reduce the burden of these diseases on our population. GENERIC NAME OF PRODUCT: Optical Pooled Screen Microscope SALIENT CHARACTERISTICS: Capable vendors must demonstrate their ability provide the best-in-class equipment and software that will allow high content and high speed performance of optical pooled screens by INDU. The key requirements of this system are needed to perform this particular imaging application. Capable vendors must demonstrate their ability to provide brand name equal equipment with the following salient characteristics. Because INDU already has two Nikon microscopes, vendors must demonstrate the equipment�s ability to integrate into existing infrastructure and allow exchange of compatible parts between the microscope systems. Key specifications of microscope: 25mm FOV (AX-R). This is required to improve throughput of the imaging, since extremely large areas on a 300 mm x 300 mm stage will be imaged during the optical pooled screen. A 25mm field of view is required to reduce the time that it takes to perform serial rounds of imaging during the ISS portion. Continuity and integration with existing systems Exchangeable optics between scopes � the INDU currently has two Nikon microscopes, therefore can exchange expensive microscope objectives between different microscopes. NIS-Elements � same software driving all of the microscopes in INDU. A central analysis computer with specialized Nikon software can therefore also be used to analyze all data regardless of which microscope the data were acquired on. AX-R unique features: 4 Channel spectral detector � tunable detector allows more than 4 channels to be defined and captured in a seamless integration/interface. Combined with the proposed 8 laser lines, spectral flexibility is maximized for the MiSeq dyes. Hybrid Galvo and resonant scanner: both achieve 25mm FOV and are matched (no resonant field cropping required) Resonant scanner max scan resolution is 2048x2048 pixels in one scan: this high pixel density allows adequate pixel sampling in an industry-best 25mm. The resonant scanner is also fast and gentle, making the AX-R uniquely equipped for high throughput applications. Continuously variable hexagonal pinhole (competitors all use square pinholes) allows more light to the detector without compromising Z rejection. This is because a hexagon more closely approximates the circular airy disc pattern. Gold plated scanning mirrors and low incident angle dichroics for minimum light loss. Fiber coupled detector provides the following advantages Isolation of detectors from heat and sources of electrical noise. Results in very sensitive low-noise detectors and small footprint of scanhead Detector is easily accessed for upgrades, field service, etc. Detectors themselves are modular. The proposed config is 3 GaAsP and 1 Multi alkali for far-red, but each individual detector can be swapped as needed. So 4 GaAsP is possible without requiring a service visit (field swappable) 8Kx8K maximum Galvo scan resolution takes full advantage of the 25mm FOV by providing 64megapixel images, which can be extremely useful for high detail imaging at low mag (translating to an extremely large FOV when lower power lenses are used). AX incorporates AI tools: Autosignal.ai � automatically determines gain and laser power for each channel based on deep learning. Single button click to optimize each channel Denoise.ai � removes detector shot noise to produce noise-free images. Will be very useful in combination with the resonant scanner to boost acquisition speeds (reduces averaging requirements) Experiment setup wizard allows users to input their dyes of choice, and the best channel setup config is selected automatically from available lasers and detectors. Users can modify these freely in advanced mode, but very useful for getting set up. DUX-VB detector (on your quote) allows continuous spectral acquisition and unmixing in continuous band mode. Water immersion dispenser 3rd party device integration (for fluidics control) Custom stage fit & control (Prior or Marzhauser) NIS-Elements and JOBS for custom acquisition routine building Includes TTL I/O control for external device triggering and programmable actions in response to incoming TTL signals Integration of the most diverse array of 3rd party devices of all major scope manufacturers. NIS-Elements supports the Lumencor Celesta light engine. FREE onsite support from the Nikon local team INSTRUCTIONS: Companies that believe they possess the capabilities to provide the required product should submit documentation of their ability to meet each of the requirements to the Contracting Officer. The capability statement must specifically address each of the requirements separately. All capability statements must also include the following: 1) company name and address; 2) point of contact; 3) phone/fax/email; 4) NAICS Code(s); 5) business size and status; 6) capability information in response to the requirement and qualifications identified in this notice; and 7) type of small business your organization classifies itself as (i.e.: Women-Owned, HUBZone, etc.). Each capability statement must include the respondent�s Unique Entity Identifier number, organization name, and address. Each response should include the technical and administrative points of contact, including names, titles, addresses, telephone numbers, and e-mail addresses. The capability statement should clearly identify the size and type of business (e.g., 8(a), HubZone, Non-Profit, Other Than Small Business, etc.), pursuant to the applicable NAICS code and any other information that may be helpful in developing or finalizing the acquisition requirements. The information submitted must be in an outline format that addresses each of the elements of the project requirement and in the capability statement paragraphs stated herein.� A cover page and an executive summary may be included but is not required.
