- This notice is being published in accordance with Federal Acquisition Regulation (FAR) Part 5.101(a)(2) requiring the dissemination of information on proposed contract actions expected to exceed $15,000, but not expected to exceed $25,000. This is a notice of intent to award a sole source contract to Mission Bio�and is not a request for competitive proposals.� This acquisition is conducted under the authority of the Federal Acquisition Regulation (FAR) Part 13�Simplified Acquisition Procedures, Subpart 13.106-1 (b) (1), Soliciting from a single source and is not expected to exceed the simplified acquisition threshold.� Contracts awarded using FAR Part 13�Simplified Acquisition Procedures and at a dollar value less than the simplified acquisition threshold are exempt from the requirements of FAR Part 6�Competition Requirements. Single-sole Source Determination: The determination by the Government to make a single-sole source�is based upon the market research conducted as prescribed in FAR Part 10�Market Research, specifically the results of the market research concluded that this order is for tapestri single cell dna sequencing offered by Mission Bio.�The Tapestri platform from Mission Bio has the following unique services. The platform utilizes droplet microfluidic technology to rapidly encapsulate, process, and profile 5,000 to 10,000 individual cells for multi-analyte detection including single-cell DNA and Protein analysis. The platform utilizes a unique two-step workflow, including (1) cell encapsulation and protease incubation to lyse cells and allow access to DNA within individual droplets and (2) barcode addition and reagent introduction for target identification and amplification. The platform allows target amplification without use of whole genome amplification methods. The platform has the capability to do both cell encapsulation and barcode/reagent addition on the same cartridge. The workflow does not take more than 4 hours of hands-on time from cell suspension to sequencing-ready libraries. The platform is compatible with an Illumina sequencing platforms MiSeq System, Illumina HiSeq 2500 System, Illumina Novaseq System. The platform can allow robust DNA detection of single-nucleotide variants (SNVs), small insertion/deletions (indels), and copy number aberrations across regions targeted by gene specific primers. The platform must be capable of operating in a mode to perform targeted DNA detection and detect protein analytes from the same cell. The platform is capable of detecting subclones present in at least 1% of the population. Mission Bio is the only company we could find that provides the single cell DNA sequencing services per the specifications required. Market research on the internet only resulted in Mission Bio as the provider for tapestry single cell DNA sequencing. A search of previous government buys on the FPDS system displayed Mission Bio as a sole source. The NIDCR investigators ordering these reagents and service want to identify copy number variations at the single cell level, in mouse tissue in which mosaic aneuploidy is suspected to be caused by genome instability. The Tapestry platform is the only one that allows to get this type of information on a large number of single cells without having to sequence the whole genome. A selection of 300 amplicons have been designed throughout the genome to allow for accurate determination of chromosome copy number variation. Product includes the complete single cell DNA workflow on 4 samples to cover training for new users (includes Core reagents, Cartridges, and DNA Oligo Pools-Myeloid panel) Quantity (1)�Tapestri Single-Cell DNA Custom Panel Oligo Pool, up to 300 amplicons - Custom targeted amplicon oligos pool supporting the processing of up to 32 samples to be used with core reagent kits. Quantity (1)�Tapestri Single-Cell DNA Training Kit, Product includes the complete single cell DNA workflow on 2 samples to cover training for new users (includes Core reagents, Cartridges, and DNA Oligo Pools-Myeloid panel) Closing Statement: This notice is not a request for competitive proposals.� However, interested parties may identify their interest and capability to respond to this notice.� Responses to this notice shall contain sufficient information to establish the interested parties� bona-fide capabilities for fulfilling the requirement and include: unit price, list price, shipping and handling costs, the delivery period after contract award, the prompt payment discount terms, the F.O.B.
