- Description
- The Laboratory of Ribonucleoprotein Biochemistry, Biochemistry and Biophysics Center, NHLBI, requires a sonication system capable of DNA and RNA shearing for Next-Generation Sequencing, chromatin shearing, protein extraction from native mammalian cells, and protein-RNA isolation from UV-crosslinked mammalian cells. This instrument must allow RNA shearing from 200 bp � 1kb and DNA shearing from 150 bp- 1 kb. It must be able to accommodate 0.2 mL, 0.65 mL, 1.5 mL, and 15 mL tubes for various applications. It must also be capable of processing up to 16 samples in parallel. The instrument must sonicate samples in a closed environment to prevent cross-contamination and aerosol formation, and samples must be rotated through the ultrasound field to ensure consistent sample processing. The sonication chamber must also be cooled to allow isothermal processing at 4 degrees C. This instrument will be used for generation of RNA, DNA, and protein samples to increase the sensitivity, accuracy and throughput of various assays. In particular, it will be used to generate reproducible samples for high-throughput sequencing analysis.
