Description
The Environmental Protection Agency (EPA) is performing a ""market search"" to identify capable and qualified sources that can provide assay kits and panels to support the evaluation of treatment-related changes in gene expression in samples collected from animal or plant species of interest to the EPA. The EPA will utilize phenotypic and genomic screening to screen samples in order to identify chemical bioactivity leading to hypotheses about chemical Mechanism of Action (MOA). In order to test MOA hypotheses, EPA requires access to a broad variety of in vitro, target-based assays and panels including binding, enzymatic and functional cellular assay formats. Target families of interest include G Protein-Coupled Receptors (GPCRs), ion channels, kinases, cytokine receptors, growth factor receptors, transporters, nuclear receptors, phosphatases, proteases, epigenetic enzymes, ubiquitin enzymes, phosphodiesterases and others. In addition, assays which evaluate the Absorption, Distribution, Metabolism, Excretion (ADME) properties of chemicals to facilitate interpretation on the in vitro results and help with extrapolation to predicted in vivo effects will also be of interest. Such assays and panels could include CYP450 induction and inhibition and transporter function. The data generated from these assays will be used to help define specific mechanisms of action along with potencies for compounds of interest to the EPA.
