- DESCRIPTION: The National Institutes of Health - NICHD intends to procure the services of a contractor to provide an OpenSPR 2-Channel Starter Pack, complete with software, Regeneration Optimization and maintenance Kits. The Lab studies molecular mechanisms of synapse development and homeostasis. They use the Drosophila neuromuscular junction as a genetic model system to study glutamatergic synapse development. They focus on the assembly and maturation of the glutamatergic synapses, in particular on the synaptic recruitment of glutamate receptors and the mechanisms that sculpt their activity-dependent stabilization at postsynaptic densities. In recent studies they uncovered a number of trans-synaptic signaling complexes important for synapse maturation and homeostatic plasticity. These multi-molecular complexes are composed of units loosely bound to each other. The attempts to capture many of these interactions have been hampered by these low binding affinities, incompatible with many biochemical assays. In addition, many of these signaling components appear to be sensitive to the extracellular Ca2+ concentrations. The entire research progress depends on understanding these Ca2+-triggered signals and their impact onto the functions enabled by the trans-synaptic macromolecular complexes. However, at this time they have no tools in the lab to capture this Ca2+ dependence and study theor role on individual protein interactions within the macromolecular signaling complexes. To be able to perform binding kinetics and affinity characterization for various trans-synaptic components in the absence of Ca2+ or at controlled concentrations, they decided to purchase a compact surface plasmon resonance (SPR) instrument, the OpenSPR from Nicoya Lifesciences inc.� The OpenSPR is the world�s only benchtop SPR instrument that can provide real time kinetic data. The OpenSPR uses a unique nano-structured sensor surface and localized SPR (LSPR) to deliver repeatable, highly sensitive kinetic data. This enables high quality, label-free interaction analysis for a fraction of the cost of all other existing SPR platforms. This technology is applicable to protein, lipids, sugars, antibodies, small molecules and even cells, offering a perfect range for examining the multimolecular trans-synaptic complexes relevant for our projects. This system also includes proprietary software to run the OpenSPR instrument and a TraceDrawer Analysis Software License, a powerful kinetic analysis software that interfaces easily with OpenSPR data. This proposed procurement is for a service for which the government intends to solicit and negotiate on a sole source basis to Nicoya LifeSciences located in Ontario, CANADA. �In order to comply with the requirements in this Special Notice, the responses should be based on our interpretation of the general requirements provided below. Note 22. The proposed contract action is for a continuation of previous services for which the Government intends to solicit and negotiate with only one source under the authority of FAR 6.302. Interested persons may identify their interest and capability to respond to the requirement or submit quotations. This notice of intent is not a request for competitive quotes. However, all quotes received within the time and date of publication of this ""Special Notice "" will be considered by the Government. A determination by the Government not to compete with this proposed contract based upon responses to this notice is solely within the discretion of the Government. Information received will normally be considered solely for the purpose of determining whether to conduct a competitive procurement. HOW TO RESPOND: In order to compete for this project, interested parties must be registered in the Central Contracting Registration and submit a price quotation by 2:00 pm, ET, July 20th 2020. Offerors may either submit one copy of the quote to MOE FILALI by mail or preferably via email. Electronic transmissions to Moe.Filali@nih.gov are preferable but must be timely. It is the responsibility of the Offeror to ensure the proposal is received by the date and time shown above.
