Description
DESCRIPTION: The National Institutes of Health - NICHD intends to procure the services of a contractor to provide a PISTON GRADIENT FRACTIONATOR - instrument and related components. Measuring the different ribosomal components in different fractions using fractionation is the most accurate and reliable method to measure the assembly of ribosomes in the cells. The Piston gradient fractionator allows for the most precise and reproducible fractionation of gradients. Lab�s rationale for sole source: ��It is now clear that RNAs are important regulators in organisms from all domains of life. In bacteria, these regulatory RNAs are typically 50-300 nt in length and are therefore denoted small RNAs (sRNAs). While not all of the sRNAs have been characterized, there have been great strides in understanding the functions of many of these regulators. The majority of the sRNAs act by base pairing with target mRNAs. Of the base pairing sRNAs with known function, a few are true antisense RNAs (asRNAs), which are encoded opposite their mRNA target and by definition have extensive complementarity with their targets. Most of the characterized base pairing sRNAs however, are encoded in trans to their target mRNAs and operate through imperfect base pairing. These sRNAs act to modulate the stability or translation of mRNAs and often control the expression of entire regulons. Most the bacteria are motile and can swim through liquid environments. One of the main structures allowing this motility is the flagellum. While transcriptional regulation of flagella genes has been studied for many years, less is known about the post-transcriptional control of the regulon by sRNAs. Our work hints that one sRNA downregulates ribosomal proteins to decrease flagella synthesis. Experiments that will enable us to measure the effect of this sRNA on the ribosomes and ribosomes assembly are extremely valuable. In addition to being extremely valuable to the project to understand the regulation of flagella synthesis, this equipment will allow us to more generally carry out ribosome profiling experiments in which the distribution of ribosomes across all transcripts is examined. Biocomp Instruments is one of the leading companies manufacturing Gradient Fractionator instruments in the US. The Biocomp system, which includes a piston gradient fractionator, Gilson fraction collection system and Triax flow cell provides all that is needed by our lab. The Piston gradient fractionator the only one that works by displacing the gradient from the top down with a special piston, producing fractions from 10 ?m to the entire gradient. Fractionation can be handled manually to recover visible bands or automatically to produce perfect gradient profiles. In addition, its compact size and its ease of use are advantageous for our lab. We have been using the Gradient Fractionator in the Dever lab with great success but need one in Building 49 to avoid transporting samples across campus.� This proposed procurement is for a service for which the government intends to negotiate with one source, BioComp Instruments located in Fredericton, NA, Canada.�� In order to comply with the requirements in this Special Notice, the responses should be based on our interpretation of the general requirements provided.� Interested persons may identify their interest and capability to respond to the requirement or submit quotations. This notice is not a request for competitive quotes. However, all quotes received within the time and date of publication of this ""Special Notice� will be considered by the Government. A determination by the Government not to compete with this proposed contract based upon responses to this notice is solely within the discretion of the Government.
