- DESCRIPTION: The National Institutes of Health - NICHD intends to procure services to synthesis and characterize novel modified peptides and create a generation and characterization of modified alcohol related peptides and generation of monoclonal antibodies specifically recognizing these peptides.� �This service is a continuation of the same work being performed by the very same vendor because of data collection integrity. This proposed procurement is for products and services for which the government intends to solicit and negotiate on a sole source basis to New England Peptides, located in Gardner, MA 01440. �In order to comply with the requirements in this Solicitation/Special Note, the responses should be based on our interpretation of the general requirements provided below. Special Note: The proposed contract action is for supplies and services for which the Government intends to solicit and negotiate with only one source under the authority of FAR 6.302. Interested persons may identify their interest and capability to respond to the requirement and/or submit quotations. This notice of intent is not a request for competitive quotes. However, all quotes received within the time and date of publication of this ""Special Notice"" will be considered by the Government. A determination by the Government not to compete with this proposed contract based upon responses to this notice is solely within the discretion of the Government. Information received will normally be considered solely for the purpose of determining whether to conduct a future competitive procurement. HOW TO RESPOND: In order to compete for this project, interested parties must be registered in the Central Contracting Registration and submit a price quotation by 4 pm, ET, July 15th, 2020. Offerors should submit one copy of the quote to MOE FILALI via email. Electronic transmissions to Moe.Filali@nih.gov are preferable but must be timely. It is the responsibility of the Offeror to ensure the proposal is received by the date and time shown above.� All questions should be directed earlier.. �Background and Lab�s Justification: The purpose of this project is to synthesis and characterize novel modified peptides and to generate monoclonal antibodies that specifically target these modified peptides.� These modified proteins were identified as potentially important contributors to alcohol-related pathologies by our lab, and specific monoclonal antibodies are needed to better characterize these emerging pathologies.� To develop an antibody to a novel target such as this requires 8 essential elements: (1) Synthesis and purification of the precursor peptide and delivery to our laboratory in a protected stable form (VENDOR completed); (2) Specific chemical modifications of the purified peptides (performed in our laboratory); (3) HPLC and mass spectrometry analyses of the modified peptides to ensure identity and purity and to quantify prior to carrier conjugation (VENDOR); (3) Deprotection and carrier conjugation of modified peptide in preparation for immunization (VENDOR); (4) Immunization and boosting with titer checks (VENDOR); (5) Antibody collection; (6) Splenocyte harvest and fusion (Vendor); (7) Clone generation and testing and selection with provision of clone supernatant and provision of promising clones to us for test (VENDOR); (8) Subcloning with delivery of supernatant (VENDOR). Several vendors offer monoclonal antibody services and we have worked with several companies in the past on antibody projects. Of these, New England Peptide (NEP) is the sole source that can complete each of the items required for successful completion of this project.� NEP routinely provides and is the leader in peptide synthesis services. Other monoclonal vendors use NEP for the synthesis and purification of protected precursor peptide (Step 1 above). To our knowledge, NEP is also the only such vendor that offers HPLC and Mass spectrometry service, and routinely provides HPLC and mass spectrometry analysis, to characterize the identity and purity of the modified peptides that will be produced by our laboratory (Step 2 above). This analytical service is essential for ensuring that a specific, well-characterized modified peptide is used for immunization and therefore that the antibodies generated are specific for the target of interest.� NEP chemists are also experienced with removal of the protective groups used by our laboratory, which is required for successful conjugation to a carrier protein.� This step is also essential for success of the project. NEP has deep experience in all aspects required for success in this project.� We have worked with them extensively in the past, and they have demonstrated that they can successfully complete these complex procedures according to our specifications. To protect potential intellectual property related to these and related novel modified peptides and antibodies, we executed a confidentiality agreement with NEP.� Therefore, NEP is the sole source that can provide these required services.
