- The USDA, Animal Health Inspection Service (APHIS) intends to award a sole source Firm Fixed Price purchase order under the statutory authority permitting Other than Full and Open Competition per statutory authority is 41U.S.C. 253(c)(1) FAR 6.302-1 to Fisher. The property or services needed are available from only one responsible qualified source at this time and no other type of property or services will satisfy these needs. This system from Bio-Rad, CFX96 Touch Real-time PCR system is the most flexible, complete, and runs the highest value solution for accurate, sensitive, and thorough quantitative PCR results. To maintain the highest standard in these evaluations, this system provides all results in minimal time and has the newest of standards required to meet today's high plex requirements. Provided by Government Scientific Source • Capability of conventional 96-well format. The capacity allows the assay of up to 96 samples in one run. • Uses typical 0.2ml tubes, strips, and plates. Will not require expensive customized reaction vessels. • Does not require the addition of detergents or external protein sources, which alters the fundamental PCR. • The base thermal cycler will be used for standard PCR. A gradient feature allows protocol optimization on the same instrument that will be used for product quantitation by real-time detection. • Cycle-by-cycle amplification plots posted in real-time throughout the run. Empowers the researcher to terminate, extend, jump forward and abbreviate runs from original program settings. • Has demonstrated simultaneous quantitation of five different targets in the same tube with various four-target multi-plex publications. This high-level multiplexing enables the end-user to gain more information from the same sample, and increase throughput. • Data points taken for all samples simultaneously, every second, throughout each data acquisition step of each cycle of the PCR reaction. Simultaneous detection of samples removes time as a variable across the 96-well format. • Multiple data points for each cycle enables the user ability finely tune their data analysis to capture the highest level of fluorescence characteristic of the chemistry employed. • LED light source for ease of maintenance and economical replacement. • Flexible gradient capacity and Melt Curve Analysis in Real-time permits gel-free optimization of annealing temps for maximum reaction efficiencies and accurate quantitation. Rapid optimization of reaction conditions are accomplished in a single run. • Allelic discrimination plots simplify identification of homozygous and heterozygous samples. • No internal reference dye required such as ROX that would obstruct a region of the usable spectrum for reporter selection. • Demonstrated dynamic range of 9 orders. Also, documented detection of low copy number of target in the presence of high copy number of housekeeping gene. • Demonstrated dynamic range of 6 orders for each target in a five-color multiplex reaction. • Raw data files are conserved. Original presentation of data based on sound defaults, but user has flexible options for resetting threshold, biasing the data points within a reading step, and assigning background. • Included one-year warranty provides on-site repair. Customer would not have to send instrument in for service. • Post-run sample designation possible. • Gene Expression Module integrated into the Maestro software. Calculates gene expression changes using choice of ΔCt or ΔΔCt method and choice of normalizing to one or multiple housekeeping genes. This high level of analysis flexibility is necessary for the calculations widely accepted in the field. • Can view multiple fluorophores in same output charts. Allows for easy comparisons between dye layers and experimental conditions This device is compatible with the existing base station and does not require any modification. Interested sources that are capable of providing the equivalent supply listed above are encouraged to submit statements of capability to the contracting office. Small business concerns are encouraged to participate. This notice of intent is NOT a request for competitive proposals, but instead a market research method to determine interested sources. A determination by the Government not to compete this proposed action is based upon responses to this notice is solely within the discretion of the Government. This is a known proprietary system and no others exist or can be found. Government Scientific Source, INC is the only known company that provides this machine. Information received will be considered solely for the purpose of determining whether conducting a competitive (i.e. non-brand name) procurement is in the best interest of the Government. Interested sources who submit data are responsible for appropriately marking information if it is proprietary in nature. Please provide past performance and references regarding your ability to provide this supply and service along with your Statement of Capability. Must be certified in this instruments application, service and of replacing parts required. (Documents of Certification may be requested.) The Government will not award a contract based on the information received, nor reimburse participants for information they provide. Information provided is strictly voluntary.
