Page 1 of 1 The Department of Veterans Affairs, Kansas City VA Health Care System, intends to negotiate on a sole source basis, pursuant to FAR 6.302-1, with Applied Biosystems for the QuantStudio ® 6 Flex (QS6) Real-Time PCR System for ongoing eye research studies. QuantStudio 6 Flex PCR System will allow the VA to quantify in vitro and in vivo real-time changes in the gene expression and gene array profiling of the co-factors and co-repressors involved in the corneal wound healing mechanisms. This equipment is the only equipment that can meet all of the VA s required salient characteristics listed below. System can be connected to the online ecosystem where instrument data can be shared and monitored among scientists and collaborators remotely. The versatility of the system allows users to multiple formats including one of three fixed block options 0.2ml Standard 96-well and 384-well. All block options have the capacity to run Fast protocols and complete runs in <30 minutes. The instrument is ready to use directly out of the box. The instrument does not require dye calibration and RNaseP Verification is provided to verify performance. The instrument is small, designed to utilize minimal bench use and fully motorized. The sample block is made up of at least 6 separate Peltier-controlled blocks (known as VeriFlex zones). The maximum temperature difference that can be programmed across the block is 25 °C. The maximum difference in temperature allowed between adjacent blocks is 5 °C. The instrument supports variable reaction volumes of 10 l to 30 l on the 0.1ml 96-well block,10 l to 100 l on the 0.2ml 96-well block, and 5-20 l on the 384-well block. The instrument can be used with multiple dyes from different companies including FAM, SYBR ® Green I, VIC ®, NED, ABY, JUN, Mustang Purple, TAMRA, Cy5, and ROX dyes. The instrument has a maximum block ramp rate of 6.5 ºC/sec. and melt curve takes place in steps varying from 0.015 °C T 3.66 °C. The instrument uses long-lasting bright white LED (Light Emitting Diode) source that has a median lifetime greater than 5 years. The instrument has a touchscreen interface that can store protocols for quick run and enables viewing of amplification plots and filtering by Sample, Target, and Task. The instrument screen allows local user accounts with PIN-protection for security. It contains on-board storage capability with memory capacity of 10GB for storing between 1000-5000 standard absolute quantification run files and is designed for maximum run protection in case of network or power outage. The instrument is designed to complete a 40 cycle real-time PCR reaction using fluorogenic 5 nuclease assay and fast chemistries in a standard 96-well plate in under 35 minutes. The instrument includes an OptiFlex System which combines 5 coupled excitation and emission filter sets to enable collection of up to 5 unique combinations of wavelengths during a single run for multiplexing on the 384-well block instrument. The instrument supports at least two homogeneous reaction chemistries, the fluorogenic 5 nuclease assay using TaqMan ® probes and the SYBR ® Green DNA binding dye chemistry. The instrument has real-time quantitative PCR installation specifications which demonstrate the ability to distinguish between 5,000 and 10,000 template copies with a 99.7% confidence level. The instrument includes a copy of free software that is designed to collect and analyze the fluorescence data for the applications of absolute quantification, relative quantification, presence/absence assays, and allelic discrimination/SNP (Single Nucleotide Polymorphism) detection. The instrument software will allow both TaqMan ® and SYBR ® chemistries in the same run by collecting different filter colors for PCR versus melt curve stages.The instrument online software allows users to batch analyze up to 500 experiments simultaneously. The instrument software includes pre-optimized run protocols for applications like Standard Curve, Relative Standard Curve, Relative Quantification Gene Expression, and SNP Genotyping with both TaqMan and SYBR Green chemistry for quick start of runs. The instrument desktop software includes a security and auditing module that facilitates 21CFR part 11 compliance. For Gene expression profiling, users can select from more than 61,000 pre-designed TaqMan ® assays representing Human, Mouse and Rat species. The vendor provides comprehensive assay design and development guidelines for real-time quantitative PCR and SNP genotyping assays. Also, the vendor offers Telephone Technical Support and Field Applications/Sales/Service Support to help solve chemistry and instrumentation problems encountered with real-time quantitative PCR and SNP genotyping. This is a notice of intent is not a request for competitive proposals; however, any responsible source who believes it is capable of meeting the requirement may submit a capability statement to the contracting office no later than Wednesday, July 31st, 12:00, EST.
