Description
NOTICE OF INTENT TO AWARD A SOLE SOURCE CONTRACT The National Institutes of Health (NIH) intends to negotiate a contract on a sole source basis with The Jackson Laboratory, Bar Harbor, ME. The purpose of the contract is to generate and deliver a knock-in mouse line carrying a unique point mutation in exon 5 of the Zbtb20 gene (NM_001164342) to the National Institute on Deafness and Other Communication Disorders (NIDCD). This mutation will change the G at cDNA position 2155 to an A (c.2155G>A), which will change the amino acid at position 719 of the encoded protein from valine to isoleucine. The modification will be made by CRISPR-Cas technology in C57BL/6J strain background and done in two phases. The first phase will include project design, consultation, and NIDCD approval, and generation of mutant mice by CRISPR technology, followed by genotyping to identify animals with the desired genotype. This phase is expected to take 14-16 weeks to complete. The second phase will involve breeding of appropriate founder animals to obtain N1 mice and is expected to take 14-16 weeks. In this phase, N1 animals will be genotyped to confirm those carrying the desired mutation, and founders will be shipped to the NIDCD at NIDCD's expense.
