- This is a combined synopsis/solicitation for commercial items prepared in accordance with the format in FAR 12.6 as supplemented with additional information included in this notice. This announcement constitutes the only solicitation and a separate written solicitation will not be issued. This solicitation number is NICHD-18-102 and is issued as a Request for Quotation (RFQ). The solicitation/contract will include all applicable provisions and clauses in effect through Federal Acquisition Circular 2005-99. The North American Industry Classification (NAICS) Code is 334516 and the business size standard is 1000 employees. However, this solicitation is not set aside for small business. This acquisition is being conducted using Simplified Acquisition Procedures in accordance with FAR Part 13. Eunice Kennedy Shriver National Institute of Child Health and Human Development (NICHD) intends to contract on a sole source basis with ProteinSimple - 3001 Orchard Parkway - San Jose, CA 95134 to procure the following: 1. PN: P100 Milo System & Starter Package: Single Cell Western System to measure cell-to-cell variation in protein expression in heterogeneous cell samples. Includes: Milo Instrument and power cable, Scout image analysis software (tweezers, two double sided antibody probing chambers and sponges), a detailed protocol and web based operational training by a ProteinSimple Applications Scientist. One-year warranty. Repairs at ProteinSimple and include parts and labor - Quantity (1) EA 2.PN: A800 On Site Milo System Training. Includes 1-day FAS time and travel - Quantity (1) EA 3.PN: 035-106 Milo Training Kit includes a Standard scWest Kit (8 chips), frozen Jurkat cell control sample, rabbit beta tubulin primary antibody and donkey anti-rabbit IgG Northern Lights 557 secondary antibody - Quantity (1) EA 4.Freight & Handling Wes - Quantity (1) EA The Milo Single-Cell Western system form ProteinSimple is ideal for measuring cell-to-cell variation in protein expression in heterogeneous cell samples. The lab studies the genetic and environmental mechanisms that regulate initial interneuron fate decisions in the embryo and prenatal mouse brain. The lab performs numerous single cell sequencing experiments that generate lists of candidate genes that may play a role in fate decisions. However, there is often a disconnect between mRNA levels obtained in these experiments and actual protein expression. The Milo Single Cell Western platform allows us to confirm the single cell transcriptome data and the protein level, which represents a significant step forward with these types of experiments. The system will allow the lab to confirm protein expression for any candidate fate determining genes we identify from single cell sequencing experiments. The ability to rely on Western-capable antibodies (rather than immunohistochemistry-grade antibodies) significantly increases the screening capability of this platform over other systems. The Milo Single-Cell Western system is a novel and complete solution for western-based protein expression measurements on single-cells. Microscale separations, quantitation and immunodetection of proteins in ~1,000 single-cells is performed in a unique scWest chip which contains a patent-protected functionalized polyacrylamide gel patterned with thousands of microwells. The system can analyze ~1000 single-cells in parallel and uses conventional western primary antibodies and standard fluorescently tagged secondary antibodies for immune probing. Multiplexing is possible using different spectrally tagged secondary antibodies or utilizing differences in target molecular weight. The immunoassay utilizes fluorescence detection for quantification of target proteins. The unique software allows the user to analyze ~1000 single-cell westerns in parallel to automatically find peaks and extract peak parameters to quantify target mobility and abundance information. The software facilitates standardization of laboratory processes and provides data in format that can be easily shared between multiple users and facilities. No other known technology combines this unique method of physical separation and immunoassay detection in single-cells. ProteinSimple is the only known company on the market that offers the technology to perform Western Blot/Protein analysis at the single cell level. ProteinSimple' s Single-Cell Western technology is protected by several issued and pending patents. The proposed contract action is for supplies or services for which the Government intends to solicit and negotiate with only one source under authority of the 41 U.S.C. 253(c) (1), FAR 6.302. The offeror must include a completed copy of the provision of FAR Clause 52.212-3, Offeror Representations and Certifications - Commercial Items with its offer. The provisions of FAR Clause 52.212-1 Instructions to Offerors - Commercial Items; FAR Clause 52.212-2, Evaluation - Commercial Items; FAR Clause 52.212-4, Contract Terms and Conditions - Commercial Items; and FAR 52.212-5, Contract Terms and Conditions Required to Implement Statutes or Executive Orders - Commercial Items - Deviation for Simplified Acquisitions applies to this acquisition. The offeror must include their Dun & Bradstreet Number (DUNS), the Taxpayer Identification Number (TIN), and the certification of business size.
