- This notice is a combined synopsis/solicitation for commercial items using Simplified Acquisition Procedures. This acquisition will be made in accordance with the format in Federal Acquisition Regulation (FAR) Subpart 12.6 "Streamlined Procedures for Evaluation and Solicitation for Commercial Items," as applicable, and as supplemented with additional information included in this notice. This announcement constitutes the only solicitation (a written solicitation will not be issued) and quotes are being requested. The Solicitation number is RFQ-RML-C-1917689 and the solicitation is issued as a Request for Quotes (RFQ). This acquisition will be processed under FAR Part 13 Simplified Acquisition Procedures (SAP). The solicitation documents and incorporated provisions and clauses are those in effect through Federal Acquisition Circular FAC 2005-99_6-15-18.pdf as of (07-03-18). Project Background The Rocky Mountain Laboratory (RML) Laboratory of Bacteriology (LB) Immunity to Pulmonary Pathogens Section elucidates the mechanism by which highly virulent pathogens, including the intracellular bacterium Francisella tularensis, evades and suppresses host immune responses to cause disease. This works utilizes in vitro cell culture of primary human and murine cells as well as an in vivo mouse model to define the molecular mechanisms by which F. tularensis and other pathogens manipulate the host immune system to cause disease and death. B. Project Description The Immunity to Pulmonary Pathogens Section performs tissue culture experiments to identify the mechanism by which pathogens manipulate host cell processes to cause disease. The use of RNA sequencing technologies is a powerful tool to understand the changes in gene expression over time, following infection. Currently, this is done in one reaction containing mixed cell populations. This provides information on gene expression in bulk cells, but does not reveal how individual cells respond. The 10X Chromium Single Cell Controller will allow us to analyze gene expression in individual cells including their relative infection status. This is a powerful technology that will greatly improve our understanding of the interface of pathogen, infected cell and uninfected by-stander cells.
