Description
PRE-SOLICITATION NOTICE OF INTENT NON-COMPETITIVE INTRODUCTION PURSUANT TO FAR Subpart 5.2-Synopses of Proposed Contract Actions, THIS IS A PRE-SOLICITATION NOTICE OF A PROPOSED CONTRACT TO ACTION. THIS IS A PRE-SOLICITATION NON-COMPETITIVE NOTICE OF INTENT TO AWARD A CONTRACT OR PURCHASE ORDER WITHOUT PROVIDING FOR FULL OR OPEN COMPETITION (INCLUDING BRAND-NAME). The National Institute on Drug Abuse (NIDA), Station Support Contracts and Simplified Acquisitions Branch (SS/SA) intends to negotiate and award a contract for a fragment analyzer. NORTH AMERICAN INDUSTRY CLASSIFICATION SYSTEM (NAICS) CODE The intended procurement is classified under NAICS code 334516 with a Size Standard of 1,000 employees. REGULATORY AUTHORITY The resultant contract will include all applicable provisions and clauses of the Federal acquisition Requlation (FAR) in effect through the Federal Acquisition Circular (FAC) 05-95, 19 Jan, 2017. STATUTORY AUTHORITY This acquisition is conducted under the authority of the Federal Acquisition Regulation (FAR) Part 13-Simplified Acquisition Procedures, Subpart 13.106-1 (b) (1), Soliciting from a single source. DESCRIPTION OF REQUIREMENT This notice of proposed acquisition is posted as an intent to award a purchase order on a non-competitive basis to: Advanced Analytical 2450 SE Oak Tree Court Ankeny, IA 50021 Advanced Anayltical is the only company that can provide this instrument based on their machine's ability to process restriction enzyme digested plasmids and PCR products and its ability to run 288 samples unattended. Project Description The NIDA Genetic Engineering and Viral Vector Core (GEVVC) serves NIDA-IRP researchers by facilitating studies of brain function under physiological and pathological conditions through developing and producing genetic tools capable of modulating and monitoring the molecules, cells and circuits in the nervous system. The NIDA GEVVC offers the following services: (1) GEVVC services that are tailored to the needs of individual IRP researchers. Services include plasmid construction, packaging of custom Adeno-associated virus (AAV) and Lentivirus (LV) vectors, genome editing consultation for the design of CRISPR reagents for targeted mutagenesis or knock-in transgenes, development of genotyping assays, and BAC recombineering; and (2) training and access to equipment necessary for molecular biology techniques, i.e. for the construction of DNA vectors, genotyping, quantification of gene expression by ddPCR and qRT-PCR, expression and purification of proteins in bacterial and mammalian cell culture. GEVVC can use the Fragment Analyzer to: (1) streamline CRISPR workflows with reliable reagent quality control and a simple and accurate screening process while simultaneously elucidating the zygosity of positive clones; (2) determine the amplicon size and concentration of PCR fragments; (3) assess the amount of supercoiled plasmid DNA in a sample; (4) reliably and accurately assess genomic DNA size, quantity and quality; (5) reliably and accurately assess messenger and small RNA size, quantity and quality; (6) quantify and qualify DNA fragments and smears through 50 kilobases; and (7) assess the size and quantity of cell-free DNA (cfDNA) which is found in serum, plasma and cerebral spinal fluid (CSF) and can be potentially used as biomarkers in various diseases states, including brain injury. Purpose and Objectives The NIDA Genetic Engineering and Viral Vector Core GEVVC needs a Fragment Analyzer which is capable of high resolution separation and quantification of DNA and RNA molecules to automate and standardize the characterization of mixed populations of nucleic acid fragments. This equipment will allow GEVVC to efficiently and cost-effectively categorize and quantify a wide range of DNA samples such as plasmids for viral vector production and PCR products amplified from CRISPR-edited cell lines and animals. This machine will also be used for similar analyses of total RNA and small RNA harvested from animal tissue and cell lines. The high resolution data generated by this machine will help investigators to decide whether or not to invest further time and resources on particular batches of samples, and should result in a substantial reduction in the amount of wasted effort. Project requirements: Fragment Analyzer with the following characteristics: 1. Technology must be centered on highly sensitive fluorescence-based, parallel capillary electrophoresis. 2. Analyze samples in as little as 15 minutes 3. View separations as they are generated and change electrophoresis times during a run if desired. 4. Direct parallel injection and separation of 12, 48, or 96 samples at once allowing the instrument to adjust throughput to fit the lab's needs. 5. The capillary array is stored in the equipment eliminating daily handling of the array. 6. Must have detection limits as low as 5pg/uL for DNA fragments and smears as low as 50pg/uL. Directly inject amplicon or restriction enzyme digestion product. 7. Must resolve a wide range of DNA and RNA fragments. Achieve as low as 3 basepair (bp) resolution with high sizing accuracy 8. High and Standard quantitative kits must include, but not be limited to: large fragment analysis for DNA 50bp-20,000bp; and RNA analysis for small RNA, microRNA, total RNA and messenger RNA. 9. Load and run up to three plates, or up to 288 samples, without human intervention. 10. Reagents are room temperature stable and no daily array handling 11. Requires only a single dilution step into the sample plate or tube strips. 12. Software packages which includes, but is not limited to: sample flagging for rapid scoring; smear analysis proving concentration, molarity, size range, average size, and CV%; ability to apply criteria to specific samples or all samples simultaneously; annotation features and customizable screen projection; and sample overlay with the ability to combine data from as many as 24 different runs Period of Performance On or before September 1, 2017. CLOSING STATEMENT This synopsis is not a request for competitive proposals. However, interested parties may identify their interest and capability to respond to this notice. Responses to this notice must include sufficient information to establish the interested parties' bona-fide capabilities of providing the product or service. The price quote shall include: unit price, list price, shipping and handling costs, delivery days after contract award, delivery terms, prompt payment discount terms, F.O.B. Point (Destination or Origin), product or catalog number(s); product description; and any other information or factors that may be considered in the award decision. Such factors may include: past performance; special features required for effective program performance; trade-in considerations; probable life of the item selected as compared with that of a comparable item; warranty considerations; maintenance availability; and environmental and energy efficiency considerations. In addition the Dun & Bradstreet Number (DUNS), the Taxpayer Identification Number (TIN), and the certification of business size must be included in the response. All offerors must have an active registration in the System for Award Management (SAM) www.sam.gov." A determination by the Government not to compete this proposed contract based upon responses to this notice is solely within the discretion of the Government. The information received will normally be considered solely for the purposes of determining whether to proceed on a non-competitive basis or to conduct a competitive procurement.
