- NOTICE OF INTENT: The USDA, ARS, SEA, intends to award a sole source procurement to 10X Genomics, Inc., 7068 Koll Center Parkway Suite 401 Pleasanton, California 94566, as they are the sole source for this type of system, pursuant to the authority of 41 U.S.C. 253(c)(1) and FAR 6.302-1, Only One Responsible Source and No Other Supplies or Services Will Satisfy agency Requirements. The award will be for a Chromium Controller and related Genome Library and Kit consumables. Specifically, the Chromium System with related consumables and software represent a unique system to discover previously inaccessible genomic information at massive rate and scale, including: phase resolved structural variants, single nucleotide variants and indels, as well as dynamic gene expression profiles from tens of thousands of individual cells. Proprietary microfluidic circuits partition samples (e.g. high molecular weight DNA, single cells) which are then barcoded using GemCode TM technology. Downstream application-specific assays generate barcoded libraries for sequencing on Illumina sequencing systems. The GemCode Gel-Beads in the Chromium system allow for barcoding DNA or RNA samples using 4,000,000 and 750,000 barcodes, respectively. This allows for individual DNA fragments to have unique barcodes in an automated fashion, where a single long strand of DNA (>100 kbp) can be fragmented and all fragments from that strand are barcoded with the same identifier. This can be done with ~1,000,000 individual reactions. When these samples are then sequenced on an Illumina DNA sequencer platform (GBRU has in-house) it is possible to leverage linked-read data which in turn allows for genome assembly in a cost efficient manner. This process can be used to also generate haplotypes by using long range phasing of whole genomes, whole exomes or targeted resequencing. The device allows for generation of exome libraries from whole cells. The Government intends to award a firm fixed price Purchase Order on or about September 01, 2016, using Simplified Acquisition Procedures. This Notice of Intent is not considered a Request for Quotation (RFQ) nor is it a Request for Proposals (RFP). No solicitation document is available and telephone requests will not be honored. Interested persons may identify their interest and capability to respond to this requirement. A determination by the Government not to compete this proposed contract based on responses to this notice is solely within the discretion of the Government. The Government will consider all responses received by 11:30 AM central time, August 31, 2016. Information received will be considered solely for determining whether to conduct a competitive procurement.
