- This is a combined synopsis/solicitation for commercial items prepared in accordance with the format in FAR 12.6 as supplemented with additional information included in this notice. This announcement constitutes the only solicitation and a separate written solicitation will not be issued. This solicitation number is NICHD-16-144 and is issued as a Request for Quotation (RFQ). The solicitation/contract will include all applicable provisions and clauses in effect through Federal Acquisition Circular 2005-89. The North American Industry Classification (NAICS) Code is 334516 and the business size standard is 1000 employees. However, this solicitation is not set aside for small business. This acquisition is being conducted using Simplified Acquisition Procedures in accordance with FAR Part 13. Eunice Kennedy Shriver National Institute of Child Health and Human Development (NICHD) intend to contract on a sole source basis with NanoTemper Technologies of Munich, Germany to procure the following items: 1. Affinity Analysis Software (unlimited license) - Quantity (1) EA 6. Product Code: MO-UPG - Update & Upgrade MO. Affinity Analysis Software, 1 year - Quantity (1) EA 7. Product Code: PACK_ML - Delivery and Setup - Quantity (1) Charge 8. Product Code: TRAIN-CUST - 1 Day Training - at customer site - Quantity (1) Charge The lab investigates the metabolism and function of brain DHA (docosahexaenoic acid) in relation to ethanol consumption. The lab have recently discovered that synaptamide (docosahexaenoylethanolamide), a metabolite of DHA, ameliorates the ethanol induced impairment of neurogenic differentiation in neural stem cells, by promoting neurite development and synaptogenesis via binding to orphan G protein-coupled receptor GPR11. To understand detailed molecular mechanisms of the synaptamide/GPR110 signaling, it is essential to purchase a biomolecular interaction system that can be used to investigate the GPCR-lipid interaction. There are many challenges associated with analyzing the GPCR-ligand interactions as GPCRs are difficult to purify with high purity and large amounts. Their stability and activity often strictly required proper liposomes or detergents. The mobilization of the GPCRs to a chip used in the SPR (surface plasmon resonance) - based technique is difficult and often causes problems due to steric hindrance at the interface. Moreover, the changes in mass or size upon the complex formation, which is important for the SPR or ITS (isothermal titration calorimetry) - based approach is expected to be very small. Currently, Monolith NT. 115 which is based on microscale thermophoresis (MST) technology is the only biomolecular interaction analyzer that can meet all the challenges faced for detecting and quantifying the GPCR-ligand interactions including the GRP110/synaptamide binding. In the MST experiments, microscopic temperature gradient is induced by an infrared laser and the directed movement of molecules is detected and quantified using covalently attached dyes (e.g. fluorescently labeled synaptamide), fluorescent fusion proteins or intrinsic fluorescence. The Monolith NT. 115 is able to detect GPCR-ligand binding events even without any surface immobilization or bulky/sensitive molecule assemblies such as liposomes and requires only very small amount of samples. NanoTemper is the only provider of the MST technology worldwide. The technology is patent protected and NanoTemper Technologies is the sole source of the equipment. The proposed contract action is for supplies or services for which the Government intends to solicit and negotiate with only one source under authority of the 41 U.S.C. 253(c) (1), FAR 6.302. The offeror must include a completed copy of the provision of FAR Clause 52.212-3, Offeror Representations and Certifications - Commercial Items with its offer. The offeror must include their Dun & Bradstreet Number (DUNS), the Taxpayer Identification Number (TIN), and the certification of business size.
