Description
The National Institute of Allergy and Infectious Diseases (NIAID) intends to negotiate on an other than full and open competition a sequel instrument from the vendor Pacific Biosciences. Sequel System including Instrument, Primary Analysis compute, Sequel Software (Instrument Control, Signal Processing, Base Calling, Quality Assessment, Alignment, Assembly, Visualization) and Installation Kit. Includes 12-month comprehensive warranty subject to Pacific Biosciences. On-site instruction of up to three (3) participants at the customer's location by a Pacific Biosciences Field Applications Specialist (FAS). Includes training kit and FAS travel-related expenses. Training includes Assay, Software and Bioinformatics. The Sequel System is designed to scale and increase as well as decrease overall system input. This was accomplished by improvements in instrument function, chemistry, polymerase, and software. The Sequel has the capability to scale throughput by adjusting the density of the ZWMs on the SMRT cell. As a result the average read length is expected to increase from 8,000-12,000 bp to lengths of >15,000bp. Also, the sequencing time per SMRT cell is adjustable per application need and ranges from 30 minutes to 6 hours. Also, there is no other instrument that meets the standards of the Sequel and there is no other vendor that can provide the Sequel System and its associated library prep polymerase and sequencing reagents. This system sequences individual DNA molecules using a proprietary sequence-by-synthesis approach. It exclusively processes SMRT Cells and reagents creating a scalable platform for sequencing and it is covered by numerous patents owned by PacBio, thus making this system unique to this particular vendor. Two such innovations are the ZMW technology that enables the isolation and real-time detection of fluorescently labeled bases incorporated at the single molecule level and the phospholinked nucleotides that produce a completely natural DNA strand. Other unique features of the Sequel include the circularized sequencing templates that allow for multiple sequence reads of a single molecule, the software pipelines that are developed specifically for analyzing long-read data, and the high quality assembly of GC high or low regions, including repeats. This is not a request for proposal. All responsible sources who are certified by Pacific Biosciences as authorized third party vendors may submit a proposal. All responsible sources may submit a response that will be considered. This will be a firm fixed price contract in accordance with the statutory authority for this sole source requirement 41 U.S.C. 253 (c) (1) as implemented by FAR 6.302-1 only one source available, no substitutions possible. A determination not to compete this requirement, based upon responses to this notice, is solely within the discretion of the Government. Offers must be submitted no later than 3:00 P.M.
