INTRODUCTION: THIS IS A PRE-SOLICITATION NON-COMPETITIVE (NOTICE OF INTENT) SYNOPSIS TO AWARD A PURCHASE ORDER WITHOUT PROVIDING FOR FULL OR OPEN COMPETITION (INCLUDING BRAND-NAME). THIS IS A NOTICE OF INTENT, NOT A REQUEST FOR A PROPOSAL. A SOLICITATION DOCUMENT WILL NOT BE ISSUED AND PROPOSALS WILL NOT BE REQUESTED. The National Heart, Lung, and Blood Institute (NHLBI) Office of Acquisition (OA) intends to negotiate and award a purchase order on a non-competitive sole source basis to ANDOR Technology, 425 Sullivan Avenue, Suite #3, South Windsor, CT 060741 to provide the following for the National Institute of Health (NIH), National Heart, Lung, and Blood Institute (NHLBI). Contract Line Items: 1. iXon Ultra 888 BV & OPTMASK-L-, Product Number: ULTRA888BV-OPTMSK-OC, Quantity: One Each; 2. LabView VI package - use with SDK3, Product Number: LABVIEW-SDK3, Quantity: One Each; 3. Comprehensive graphical user interface (Windows XP, Vista, 7, 8) for camera control, data acquisition and processing, Product Number: SOLIS (1), Quantity: One Each; and 4. Shipping. Background : The National Institute of Health (NIH) is the nation's leading medical research agency and the primary Federal agency seeking fundamental knowledge about the nature and behavior of living systems and the application of that knowledge to enhance health, lengthen life, and reduce illness and disability.. The National Heart, Lung, and Blood Institute (NHLBI) provides global leadership for a research, training, and education program to promote the prevention and treatment of heart, lung, and blood diseases and enhance the health of all individuals so that they can live longer and more fulfilling lives. The Biochemistry and Biophysics Center carries out research that brings chemical and physical approaches to the study of biological problems. The principal investigators of the Center focus on topics that range from DNA transcription to cellular degeneration. To elucidate the mechanisms involved in these diverse processes, the investigators develop instruments and techniques to resolve, quantify, model, manipulate, and simulate biological mechanisms at molecular and cellular levels. The focus of Center research is to develop both experimental and theoretical models of biomolecular structure, and use these models to discover the link between the structure, function, and regulation of biologically active molecules and processes. The Laboratory for Center for Biochemistry and Biophysics is constructing an instrument that combines single-molecule manipulation and fluorescence microscopy in a multiplexed format. These experiments require a large field of view, single photon sensitivity, and fast sampling rates to resolve changes in single molecules in real time. This solicitation is to procure Andor iXon Ultra 888 EMCCD camera system, which we will use in several specific experiments: 1. The Government will track the 3D trajectories of fluorescent Nano diamonds tethered to DNA molecules or proteins and measure the discrete changes in DNA extension and twist, or protein translocation and rotation, that coincide with duplex formation and catalytic activity steps of DNA associated proteins. 2. The Government will measure the binding of human topoisomerase II to supercoiled DNA, which is a crucial measurement for establishing mechanisms of the top II poisons that constitute a major class of antibiotic and chemotherapy drugs. 3. The Government will use fluorescence to measure free energy changes that cannot otherwise be resolved by existing single-molecule manipulation techniques. For example, fluorescence resonant energy transfer to measure the equilibrium constant (the fractional occupancy of two states). By performing this measurement as a function of applied forces in the sub-piconewton range, the scientific investigators will determine the spatial separation between the two states with sub-nanometer precision. This will greatly extend the capability to resolve structural changes in single biological molecules in a minimally perturbative manner. The Andor iXon Ultra 888 EMCCD camera system will allow investigators to carry out these and many other specific research activities that would not otherwise be possible. Purpose and Objectives : The Laboratory of Single Molecule Biophysics uses single-molecule techniques to elucidate the molecular mechanisms of enzymatic activity and inhibition by chemotherapeutic agents, thus fulfilling the mission of the National Institutes of Health, which is to "seek fundamental knowledge about the nature and behavior of living systems." The instruments used for these investigations are built in house and tailored for specific and demanding measurement requirements. The Andor iXon Ultra 888 EMCCD camera system will be used to add multiplexed single-molecule fluorescence measurement capabilities to an existing multiplexed magnetic tweezers single-molecule manipulation instrument. The Andor iXon Ultra 888 EMCCD camera system will form an integral component of a custom multiplexed single-molecule measurement apparatus. Period of Performance : Issued upon Award. Regulatory Authority: This acquisition is conducted under the authority of the Federal Acquisition Regulations (FAR) Subpart 13.106-1(b) Soliciting from a single source, only one responsible source and no other supplies or services will satisfy agency requirements. Additional Information : Industry Classification (NAICS) Code is 334516, Analytical Laboratory Instrument Manufacturing and the Small Business Size Standard is 500 Employees. The acquisition is being conducted under FAR Part 13, simplified acquisition procedures, therefore the requirements of FAR Part 6 B Competitive Requirements are not applicable (FAR Part 6.001). The resultant Contract will include all applicable provisions and clauses in effect through the Federal Acquisition Circular (FAC) 05-83 (August 5, 2015). This requirement is under the Simplified Acquisition Threshold of $150,000.00. This synopsis is not a request for competitive proposals. However, interested parties may identify their interest and capability to respond to this notice. Responses to this notice shall contain sufficient information to establish the interested parties' bona-fide capabilities for fulfilling the requirement and include: unit price, list price, shipping and handling costs, the delivery period after contract award, the prompt payment discount terms, the F.O.B. Point (Destination or Origin), the Dun & Bradstreet Number (DUNS), the Taxpayer Identification Number (TIN), and the certification of business size. All offerors must have an active registration in the System for Award Management (SAM) www.sam.gov." A determination by the Government not to compete this proposed contract based upon responses to this notice is solely within the discretion of the Government. The information received will normally be considered solely for the purposes of determining whether to proceed on a non-competitive basis or to conduct a competitive procurement.
