Description
This is a combined synopsis/solicitation for commercial items prepared in accordance with the format in Subpart 12.6, as supplemented with additional information included in this notice. This announcement constitutes the only solicitation; proposals are being requested and a written solicitation will not be issued. Brand Name or Equal. Minimum Essential Characteristics for KAPA (Brand Name or Equal) KAPA HiFi HotStart ReadyMix 500 x 25ul Rxn (MANF#KK2602): Used for high-fidelity PCR of a wide range of targets and fragment sizes; specifically low quality DNA samples. High fidelity PCR is used to selectively enrich library fragments carrying appropriate adaptor sequences and to amplify the amount of DNA prior to sequencing. Shall not require accessory protein domains. Shall have proven ability to amplify difficult templates (e.g AT- and GC-rich). Shall routinely amplify targets up to 15 kb from genomic DNA or 20 kb from less complex targets. Shall have proven error rate 100X lower than wild-type Taq DNA polymerase. Master Mix shall contain HiFi HotStart DNA Polymerase (1 U per 50 l reaction) in a reaction buffer containing dNTPs (0.3 mM of each dNTP at 1X), MgCl2 (2.5 mM at 1X) and stabilizers. Supplied DNA Polymerase must have proven error rate of 1 error per 3.6 x 106 nucleotides incorporated. DNA Polymerase must have 5'- greater than 3' polymerase and 3'- greater than 5' exonuclease (proofreading) activity, but no 5'- greater than 3' exonuclease activity. Compatible with sequencing on Illumina platforms. KAPA HiFi HotStart Uracil+ ReadyMix 250 x 50uL Rxn (MANF#KK2802): Used for high-fidelity PCR of a wide range of targets and fragment sizes; specifically damaged DNA samples. High fidelity PCR is used to selectively enrich library fragments carrying appropriate adaptor sequences and to amplify the amount of DNA prior to sequencing. Shall contain DNA polymerase specifically engineered to tolerate uracil residues and sequence through DNA damage Shall have proven compatibility for applications employing bisulfite DNA conversion, producing low concentrations of AT-rich DNA Shall be suitable for methylation-specific PCR Shall have proven ability for use with UDG to prevent false positives due to carryover amplicon contamination. Master Mix shall contain all components for PCR, except primers and template. KAPA Library Quantification Kit - Illumina/ROX Low (MANF#KK4873): A qPCR-based quantification of NGS libraries prior to pooling for capture or flow cell amplification on the MiSeq. DNA Polymerase contained in the qPCR Master Mix shall amplify GC- and AT-rich DNA fragments of different lengths with similar efficiency, allowing for accurate qPCR-based quantification of NGS libraries. Shall contain a pre-diluted set of DNA Standards (1-6) representing a 10-fold dilution series (20 pM to 0.0002 pM) of a linear, 452 bp template. Standards must be quality-controlled to ensure lot-to-lot consistency and avoid data drift over time. Shall contain optimized primer premixes to ensure optimal PCR efficiency. Shall contain 50X ROX Low qPCR master mix For use on Life Technologies Model 7500 qPCR instrument Compatible with sequencing on Illumina platforms KAPA Hyper Prep Kit for Illumina with Library Amplification Primer Mix (10X) - 96 Rxn (MANF#KK8504); A kit for the construction of DNA libraries for Illumina sequencing in 2-3 hrs, using low quality sample DNA Shall have proven low duplication rates and improved coverage Shall generate high-quality libraries from 250 ng FFPE DNA or less Shall allow for use of higher adaptor concentrations to improve library diversity and reduce duplication rates Input sample amount shall be between 1 ng-1 g Shall be compatible with whole genome sequencing, targeted sequencing, PCR-free workflows, FFPE and low input samples Shall employ one-tube chemistry to improve library yield and quality for FFPE and low-input libraries. Shall contain separate, concentrated enzyme formulations and reaction buffers for end repair and A-tailing as well as ligation. Shall be compatible with the MiSeq Illumina sequencing instrument KAPA HyperPlus Kit With Library Amplification 96 Rxn (MANF#KK8514): A kit for the fragmentation and amplification of DNA libraries for Illumina sequencing in a single tube, using high quality DNA samples. Shall utilize enzymatic fragmentation Average DNA fragmentation and library prep shall be around 2.5 hours Shall allow for flexible DNA sample input; from 1 ng - 1 g Shall support PCR-free workflows Kit shall contain KAPA HiFi HotStart DNA Polymerase Shall be compatible with the MiSeq Illumina sequencing instrument PROPOSAL DUE DATE: No Later Than (NLT): 14 August 2015 at 4:00 PM Eastern Time. Price quotes must be emailed to Darrell Beaver at darrell.l.beaver4.civ@mail.mil.
