- Minimal essential components for Upright Multiphoton Confocal Microscope (for wide-field imaging in fixed and live tissues). The Johnson lab is purchasing an upright multiphoton microscope system for: (1) real-time imaging of fluorescent protein reporters in transgenic mouse brains in vivo using intracranial window technology; and (2) endogenous fluorophore expression or immuno-based fluorescent labels in fixed and cleared brain slices using wide-field objectives. A fully integrated, turn-key system with all lasers, waveguides, software (acquisition and analysis), hardware and imaging systems for these capabilities is requested. This system should have the following characteristics: a.Motorized microscope with automated X, Y, Z stage and programmable multi-point, time-lapse imaging. b.Adjustable objective-to-stage clearance of 60 mm (max) for insertion of small animal stereotaxic frame on stage c.Stage must be able to accommodate small animal stereotaxic frame of 17 in L x 10 in W d.Capable of imaging two colors in in vivo brain tissues to a depth of 750 m. e.10x, 25x and 40x objectives for real-time imaging in in vivo brain tissues f.Capable of imaging two colors in uncleared and fixed brain tissues to a depth of 750 m. g.10x, 25x and 40x objectives for imaging uncleared and fixed brain h.Capable of imaging two colors in cleared brain tissues to a depth of at least 4 mm i.Wide-field 2-mm diameter 25x objectives for viewing in vivo brain tissue at varying depths j.Wide-field 4-mm and 8-mm diameter 25x objectives for viewing cleared brain slices (using CLARITY or other high-fidelity clearing methods to reduce tissue scattering) k.Live-imaging capabilities of greater than 512x512 dpi with speeds of greater than 25 frames/second while simultaneously conducting 458 nm stimulation of neurophotonic proteins/compounds. l.Capable of live imaging of greater than 512x16 dpi with speeds of greater than 350 frames/second while simultaneously conducting 458 nm stimulation of neurophotonic proteins/compounds. m.Multiphoton non-descanned detectors to enable imaging of at least two fluorophores (e.g., GFP and RFP/mCherry) n.Includes at least one 2-channel GaAsP detector to increase the dynamic range of the detection signal o.Data acquisition should include a computer with the following minimum characteristics: windows 7 (64 bit) with a 3.6 GHz quad-core processor, at least 16 GB RAM, a 2 TB hard drive, a 240 MB solid-state hard drive, a 30 quote mark high definition monitor and software for controlling all facets of the system operation. p.Data analysis system should include a second computer with the following minimum characteristics: a 3.6 GHz quad-core processor, 8 GB RAM, graphics card 1 GB DDR3 total frame rate buffer with a memory bandwidth of 25.6 GB/sec, a 2 TB hard drive, a 30 quote mark high definition monitor and software for data review and manipulation. q.Software should include non-expiring licenses to utilize all possible microscope/software functions, including single and multi-photon imaging, multi-area tiling, auto-focus, 3-D model generation and multiple area time-lapse analysis. r.Tunable laser allows multiphoton excitation from at least 690 - 1000 nm, with pulse lengths of under 100 femtoseconds and negative chirp to increase deep scanning brightness and excitation efficiency. s.Integrated auto tuning of laser power, laser diameter and detector sensitivity for brightness consistency at multiple tissue depths t.Epifluorescent light source and filter sets to visualize FITC, TRITC and mCherry wavelengths u.Vibration-free table with pneumatic isolators
