- This is a combined synopsis/solicitation for commercial items prepared in accordance with the format in FAR 12.6 as supplemented with additional information included in this notice. This announcement constitutes the only solicitation and a separate written solicitation will not be issued. This solicitation number is NIHLM2014201 and is issued as a Request for Quotation (RFQ). The solicitation /contract will include all applicable provisions and clauses in effect through Federal Acquisition Circular 2005-72. The North American Industry Classification (NAICS) Code is 541380 with a size standard of $14000000 Million. This acquisition is being conducted using Simplified Acquisition Procedures in accordance with FAR Part 13. The National Institutes of Health (NIH), National Institute of Digestive, Diabetes & Kidney Diseases (NIDDK) has a requirement to procure NGS sequencing of five DRD genes.. The objective of this project is to perform next generation sequencing of the five human DRD genes and exon 8 of ANKK1 on the Illumina HiSeq TM platform for 34 samples. Gene sequences will be captured from genomic DNA and sequenced, and then analyzed for variants (SNPs and indels). Next generation sequencing of the five human DRD genes and exon 8 of ANKK1 from 34 human buccal samples will be performed. Gene sequences will be captured and sequenced and then analyzed for variants. Vendor shall complete the required analyses with the fastest turnaround time. NIDDK currently has 34 buccal samples for analysis. Thirty four (34) human buccal swab samples collected with the BuccalAmp DNA extraction kit will be shipped to vendor. Total DNA will be extracted from each device using the QuickExtract DNA extraction solution as per manufacturer's instructions. Extracted DNA will be evaluated by NanoDrop spectroscopy, agarose gel electrophoresis and Qubit. DNA will be used for target enrichment using the CATCH-Seq Kit according to the manufacturer's protocol, and indexed libraries will be generated for each sample. Appropriate quality control analysis will be performed at every step, and the libraries will be quantified with Qubit and/or the 2100 Bioanalyzer. Libraries will be pooled in equimolar ratio, and the final pool re-quantified with Qubit. 6-8 µmol of the pooled library will be loaded onto HiSeq2500 TM rapid run flow cell to generate high quality paired end reads (2x150). Sequence data to an average depth of coverage of 400X will be generated for each sample. Raw Illumina reads will be converted into.fastq format. Sequence reads will be aligned with the reference sequences for the DRD genes. Variants (SNPs and indels) will be identified and reported. The offeror must include a completed copy of the following provisions: 1) FAR Clause 52.212-1 Instructions to Offerors - Commercial items; 2) FAR Clause 52.212-2, Evaluation - Commercial Items. As stated in FAR Clause 52.212-2 (a) The Government will award a contract resulting from this solicitation to the responsible offeror whose offer conforming to the solicitation will be advantageous to the Government, price and other factors considered. The following factors will be used equally to evaluate offers: Technical Evaluation, Price, and Past Performance. Note: Past Performance Information: Vendors must submit a listing of the most recent contracts/awards (minimum of 3) which demonstrate similar work in nature to this Solicitation. Contracts/awards may include those entered with the Federal Government, state and local governments and commercial concerns. Include the following information for each contract or subcontract: 1. The Dun and Bradstreet Number (DUNS), the Taxpayer Identification Number (TIN) and the certification of business size shall be included.
