This announcement constitutes the only solicitation; proposals are being requested and a written solicitation will not be issued. (ii) The solicitation number is NHLBI-CSB-HL-2014-172-JEF and the solicitation is issued as a request for proposal (RFP). Offerors shall submit their proposals no later than 12:00 p.m. Eastern Standard Time(EST) on Monday, July 21, 2014. (iii) The solicitation document, the incorporated provisions and clauses are those in the Federal Acquisition Circular 2005-74, effective July 1, 2014. (iv) The associated North American Industry Classification System (NAICS) code for this procurement is 334516 and the small business size standard is 500 employees. The acquisition is being conducted in accordance with the procedures of FAR Part 12 and FAR Part 13. PURPOSE: The purpose of this requirement is to perform experiments for the National Heart, Lung, and Blood Institute (NHLBI)/Division of Intramural Research (DIR) that require a Raman system which is sensitive enough to detect small signal changes produced by protein conformational changes. The sensitivity of the system will determine the limits of lab experiments and has been an important consideration since individual liposomes with relatively few proteins bound on the surface will be targeted. The lab expects the quality of the CCD camera will be the most important consideration for these experiments. The lab requires a system which can couple to the existing Olympus Ix71 microscope so correlative microscopy can be done as well as incorporating other tools such as DIC and confocal capabilities. (v) Background: The National Heart, Lung, and Blood Institute (NHLBI), Division of Intramural Research (DIR) lab has been focused on the molecular mechanisms of amyloid formation, protein conformational structure, and protein-membrane interactions. A range of spectroscopic techniques are used including circular dichroism spectroscopy, time-resolved fluorescence, and FRET to characterize protein conformations. Raman spectroscopy will provide the ability to characterize changes in protein conformation without the use of fluorescent labels which potentially create artifacts. Specifically, a Raman system will allow the lab to characterize alpha-synuclein amyloid conformations in vitro. After determining the characteristic signals for amyloid it is expected to be able to identify and monitor label-free amyloid formation in live cells. NHLBI/DIR is also focused on protein-membrane interactions of alpha-synuclein and apolipoproteins. Fluorescence imaging techniques are used to directly measure the interaction between membranes and protein. By coupling our Raman system to the existing Ix71 Olympus microscope the lab will be able to correlate fluorescence imaging with Raman signals. The Raman capabilities will be used to pursue label-free methods which will give more flexibility in designing the experiments and be more closely aligned to native systems. CONTRACTOR REQUIREMENTS: I. The contractor shall provide the following requirements for the Raman System: 1) Focal Length: Minimum 300 mm 2) Aperture: Minimum f/4 3) Resolution: Minimum 5 cm-1 4) Grating Turret: Computer controlled and minimum 3 grating options 5) Available Grating Sizes: 600, 1200, and 1800 gr/mm 6) Entrance slit: Covers 0-2 mm range and minimum 10 µm step resolution. 7) Camera shutter: Computer controlled 8) Side entrance slit: Adaptable to a fiber coupled light source with motorized mirror 9) Software: Included 10) Camera - Minimum Dynamic Range - 16 bit 11) Camera - Maximum Dark Current 2e-/pixel/hour 12) Camera - Liquid Nitrogen Cooled (minimum 24 hours volume capacity) or equivalent cooling system. 13) Camera - Back Illuminated Deep Depletion CCD Chip 14) Camera - Uniform spectral response in the range of 500-900 nm 15) Camera - Fringe suppression in the spectral range of 500-900 nm. 16) Camera - Quantum efficiency of ~90% in the spectral range of 500-900 nm. 17) Camera - CCD resolution minimum 1000 x 100. 18) Physical Dimensions: Maximum footprint of length 35" and width 32" including the camera and adapters which connect the system to the Olympus Ix71 microscope. II. The contractor shall deliver the equipment as defined in the scheduled delivery date and location. (vi) Delivery schedule will be negotiated upon award, but for purposes of proposing this delivery, please assume Net 30 days ARO Delivery location is Bethesda, MD 20892. (vii) The provision FAR clause 52.212-1, Instructions to Offerors - Commercial Items, applies to this acquisition and is hereby incorporated by reference. (viii) The provision at FAR clause 52.212-2, Evaluation - Commercial Items, applies to this acquisition and is hereby incorporated by reference. An award will be made to the lowest cost technically acceptable. Multiple awards will not be made. Evaluation and award procedures in FAR 13.106 apply. (ix) In accordance with FAR Clause 52.212-3, Offeror Representations and Certifications - Commercial Items, Offerors must complete annual representations and certifications on-line at https://www.sam.gov/portal/public/SAM/. (x) FAR clause at 52.212-4, Contract Terms and Conditions - Commercial Items, applies to this acquisition. (xii) The Defense Priorities and Allocations System (DPAS) and assigned rating does not apply to this acquisition.
