Description
The National Cancer Institute (NCI), Center for Cancer Research (CCR), plans to procure on a sole source basis a Hydrogen/Deuterium Exchange (H/DX) Liquid Handling System from LEAP Technologies, P.O. Box 969, Carrboro, NC 27510. This acquisition will be processed under FAR Part 12 - Acquisition for Commercial Items and will be made pursuant to the authority in FAR 13.106-1(b)(1) using simplified acquisition procedures for commercial acquisitions. The North American Industry Classification System code is 334516 and the business size standard is 500 employees. Only one award will be made as a result of this solicitation. This will be awarded as a firm fixed price type contract. It has been determined there are no opportunities to acquire green products or services for this procurement. Delivery of the instrument shall occur within 4-8 weeks after receipt of order (ARO). The Collaborative Protein Technology Resource Mass Spectrometry Facility performs cutting-edge mass spectrometry experiments in collaboration with researchers within the Center for Cancer Research at the National Cancer Institute. These studies into protein function and regulation are critical to advance cancer research. As the proteins are commonly mis-regulated in tumors, elucidating the function and regulation of proteins in normal cells and how they are altered in tumor cells is critical for prevention of tumor formation and growth. One aspect of these studies is the ability to use hydrogen/deuterium exchange mass spectrometry (H/DX-MS) to examine changes in protein flexibility related to environment (e.g., pH) or interaction with ligands, co-factors, other proteins, or inhibitors. The technique relies on the accurate measurement of the degree of labeling of the protein by deuterated hydrogen during a precisely measured labeling interval. Temperature control is also critical for the success of the experiment. H/DX-MS is difficult to perform manually due to the demands for precise timing and temperature control, as well as the need to reproduce exactly the same experimental conditions. Use of robotic liquid-handling systems solves both of these challenges and further improves experimental throughput. The purpose of this acquisition is to procure a liquid handling robot for automation of Hydrogen-Deuterium Exchange mass spectrometry experiments. This new system will allow CCR researchers to explore conformational changes in proteins that occur in different solvent environments (e.g., pH), in the presence of ligands or interactors, or upon small-molecule inhibition, providing novel insight into protein regulation and function. The H/DX Liquid Handling System must include a wide range of features, including: - Temperature-Controlled Drawers: • Drawers can be independently controlled so that each box can be held at a different temperature for storage of samples and reagents or for performing hydrogen/deuterium exchange reactions. • Each drawer holds two plates and offers temperature control from 1 °C - 40 °C. - Temperature-Controlled Chromatography Module: • The chromatography module has temperature control from -4 °C - 40 °C. • Comprise a pepsin digestion column; a reverse-phase separation column and trap; two column switching valves sustaining up to 15,000 psi; and an LC injection valve. - LEAP Shell Control Software: • Allows full control of each parameter and device within the system. • Enables scheduling of experiments to maximize throughput and minimize mass spectrometer downtime. • No limit to the number of protein samples which can be handled in a single experiment. • Run-time display of the measured labeling time next to the requested labeling time (accuracy is within 1 second), as well as the status of each sample as it progresses through the experiment, e.g. "Labeling", "quenched", "injected". • Can be integrated with Xcalibur (for control of the mass spectrometer). - Contractor shall provide a 12 month warranty for all modules and parts. - Contractor shall provide installation, training, and technical support. LEAP Technologies is the original manufacturer and sole supplier of the H/DX Liquid Handling System. This system combines robust and precise liquid-handling robotics with precision temperature control and proprietary scheduling software. These features are essential for reproducible execution of hydrogen/deuterium exchange mass spectrometry experiments. Therefore, to the knowledge of the Contracting Officer's Representative, Leap Technologies is the only source known that can meet the Government's needs. This notice is not a request for competitive quotation. However, if any interested party, especially small business believes it can meet the above requirement, it may submit a proposal or quote for the Government to consider. The response and any other information furnished must be in writing and must contain material in sufficient detail to allow NCI to determine if the party can perform the requirement. Responses must be received in the contracting office by 4:00 PM EST, on July 24, 2014. All responses and questions must be in writing and faxed to 240-276-5399 or emailed to Reyes Rodriguez, Contracting Specialist via electronic mail at reyes.rodriguez@nih.gov. A determination by the Government not to compete this proposed requirement based upon responses to this notice is solely within the discretion of the Government. Information received will be considered solely for the purpose of determining whether to conduct a competitive procurement. No collect calls will be accepted. In order to receive an award, contractors must be registered and have valid certification in the Central Contractor Registration (CCR) and the Online Representations and Certifications Applications (ORCA) through sam.gov.
