- This is a combined synopsis/solicitation for commercial items prepared in accordance with the format in Subpart 12.6, as supplemented with additional information included in this notice. This announcement constitutes the only solicitation; proposals are being requested and a written solicitation will not be issued. Minimum Essential Characteristics (MECs) 1.-Required for the routine detection of botulinum neurotoxin (BoNT) serotypes A B, C, D, E, F and G. The assay must have the ability to measure accurately BoNT's proteolytic cleavage of natural BoNT substrates - SNAP25 or VAMP2 - in a sensitive, FRET-based, mix and read format and using most standard fluorescent plate readers. 2.-The FRET-based nature of the assays allows for real-time detection of BoNT proteolytic activity enabling the determination of kinetic constants and the determination of enzymatic activity. 3.-Assay must have picomolar to fentomolar sensitivity. 4.-Assay must contain a reporter that consists of residues 141-206 (66-MER) of SNAP-25, the naturally occurring substrate of BoNT/A and E. The substrate must be sandwiched between two fluorescent proteins, a cyan fluorescent protein (CFP) derivative, and a yellow fluorescent protein (YFP) derivative. 5.-The CFP and YFP moieties on the 66-MER (SNAP-25 derivative) must form a donor-acceptor F rster Resonance Energy Transfer (FRET) pair. 6.-Reporter must have an excitation wavelength of 435 nm and an emission wavelength of 527 nm for SNAP-25 cleavage detection. 7.-Assay must contain a reporter that consists of residues 33-94 (62-MER) of synaptobrevin, the naturally occurring substrate of BoNT/B/D/F/and G. The substrate must be sandwiched between two fluorescent proteins, a cyan fluorescent protein (CFP) derivative, and a yellow fluorescent protein (YFP) derivative. 8.-The CFP and YFP moieties on the 62-MER (synaptobrevin derivative) must form a donor-acceptor F rster Resonance Energy Transfer (FRET) pair. 9.-Reporter must have an excitation wavelength of 435 nm and an emission wavelength of 527 nm for synaptobrevin cleavage detection. 10.-Assay must include a viable reaction (HEPES based) buffer system that is favorable for both Light chain and holotoxin studies. 11.-Must include 30 assay kits that test for BoNT, A/E (SNAP25 cleavage) and 20 assay kits that test for BoNT/B/D/F/and G (synaptobrevin cleavage). 12.-Must include shipping and handling costs in order.
