INTRODUCTION : THIS IS A PRE-SOLICITATION NON-COMPETITIVE (NOTICE OF INTENT) SYNOPSIS TO AWARD A CONTRACT OR PURCHASE ORDER WITHOUT PROVIDING FOR FULL OR OPEN COMPETITION (INCLUDING BRAND-NAME). The National Heart, Lung, and Blood Institute (NHLBI) Office of Acquisition (OA) on behalf of the National Institute of Arthritis and Musculoskeletal and Skin Diseases (NIAMS), intends to negotiate and award a purchase order on a noncompetitive sole source basis to Illumina Inc., 9885 Towne Centre Drive, San Diego, California 92121-1975 to procure the following: (BRAND-NAME: ILLUMINA MISEQ SEQUENCING SYSTEM) with trade-in. Procurement : 1. MiSeq System, Catalog Number: SY-410-1003, Quantity: One (1) Each. (Trade-In for Ion Torrent Personal Genome Machine and Precision Dell Computer, Model: 508U00, Serial Number: 11C081101. 2. MiSeq Reagent Nano Kit, v2 (300 cycles), Catalog Number: MS-103-1001, Quantity: Five (5). 3. Nextera XT DNA Sample Preparation Kit (24 Samples), Catalog Number: FC-131-1024, Quantity: Three (3). 4. Nextera XT Index Kit (24 indices, 96 samples), Catalog Number: FC-131-1001, Quantity: three (3). 5. Shipping and Insurance. Background: The National Institutes of Health (NIH) is the nation's leading medical research agency and the primary Federal agency conducting and supporting medical discoveries that improve people's health and save lives. The National Institute of Arthritis and Musculoskeletal and Skin Diseases (NIAMS) are a part of the National Institutes of Health (NIH), the world's premier biomedical research agency. NIAMS supports research into the causes, treatment, and prevention of arthritis and musculoskeletal and skin diseases; the training of basic and clinical scientists to carry out this research; and the dissemination of information on research progress in these diseases. The NIH Center for Regenerative Medicine (NIH-CRM) is interested in evaluating the activity and specificity of TALEN and CRISPR designer nucleases, which are the key technologies for high-efficiency genome editing in human stem cells. The quickest and most accurate evaluation of assembled TALEN and CRISPR is based on analyzing the sequences and measuring the frequencies of TALEN/CRISPR-generated mutations (indels mostly). NIAMS high-throughput research plan, Next-Gen sequencing is much more efficient and economical than conventional Sanger sequencing because enormous amounts of targeted DNA fragments need to be sequenced. With MiSeq and its compatible Nextera XT DNA sample preparation kits that are designed for fastest and easiest preparation of PCR amplicons and small genome, we can pool 24 to 96 samples together using included index primers to fit in one run to achieve 300Mb output, after high-throughput nucleofection and genomic PCR. The sequencing output will show both sequence and frequency of each mutation generated by assembled TALEN/CRISPR. The purchase of this machine with its supplies will enable us to validate hundreds of TALEN/CRISPR we designed and assembled for genome engineering of human stem cells. Delivery: Delivery will be made upon award. REGULATORY AUTHORITY: This acquisition is conducted under the authority of the Federal Acquisition Regulations (FAR) Subpart 13.106-1(b) Soliciting from a single source, only one responsible source and no other supplies or services will satisfy agency requirements. ADDITIONAL INFORMATION: Industry Classification (NAICS) Code is 334516, Analytical Laboratory Instrument Manufacturing and the Small Business Size Standard is 500 Employees. The acquisition is being conducted under FAR Part 13, simplified acquisition procedures, therefore the requirements of FAR Part 6 B Competitive Requirements are not applicable (FAR Part 6.001). The resultant Contract will include all applicable provisions and clauses in effect through the Federal Acquisition Circular (FAC) 05-69 (September 3, 2013). This requirement is under the SAT of $150,000.00. This synopsis is not a request for competitive proposals. However, interested parties may identify their interest and capability to respond to this notice. Responses to this notice shall contain sufficient information to establish the interested parties' bona-fide capabilities for fulfilling the requirement and include: unit price, list price, shipping and handling costs, the delivery period after contract award, the prompt payment discount terms, the F.O.B. Point (Destination or Origin), the Dun & Bradstreet Number (DUNS), the Taxpayer Identification Number (TIN), and the certification of business size. All offerors must have an active registration in the System for Award Management (SAM) www.sam.gov." A determination by the Government not to compete this proposed contract based upon responses to this notice is solely within the discretion of the Government. The information received will normally be considered solely for the purposes of determining whether to proceed on a non-competitive basis or to conduct a competitive procurement.
