Description The National Institutes of Health (NIH), National Cancer Institute (NCI), Division of the Center for Cancer Research (CCR) Laboratory of Biochemistry and Molecular Biology (LBMB) intends to procure the Brand Name AKTA Pure M1 Chromatography System. This is a combined synopsis/solicitation for commercial items, prepared in accordance with format in FAR 12.6 as supplemented with additional information included in this notice. This announcement constitutes the only solicitation and a separate written solicitation will not be issued. This solicitation: No. RFQ-NCI-¬¬¬¬¬¬¬¬¬¬¬130105-TG includes all applicable provisions and clauses in effect through FAR FAC 2005-69 (August 2013) simplified procedures for commercial items. The North American Industry Classification System code is 334516 and the business size standard is 500 employees. Only one award will be made as a result of this solicitation. This will be awarded as a firm fixed price type contract. I. Description of Requirement Laboratories within the Laboratory of Biochemistry and Molecular Biology (LBMB) investigate many aspects of chromosome biology, including heterochromatin formation, chromosome segregation, regulation of genome stability, and maintenance of telomeres. Particularly relevant to this acquisition is the Laboratory of Biochemistry and Molecular Biology (LBMB) has need of the AKTA pure M1 system to purify antibodies and proteins that will be used in the Xenopus Egg extract system, human cancer cells and in vitro to study the process of cell division. LBMB has optimized protocols and successfully purified samples and generated data using this system. One of the columns used extensively for the purification of samples is a pre- packed Superdex column for high-resolution separation of biomolecules by size (gel filtration). It is critical that the work be performed using the same system (and columns) to be able to compare to previous data collected and without jeopardizing the quality of the purified samples. Parts required: 1) AKTA pure M1: system pumps (up to 25ml/min, 20MPa); gradient mixer; injection valve; simultaneous, triple wavelength UV detector, 190-700nm; conductivity detector; inlet valve V9-IAB; outlet valve V9-Os Digital copies of user manuals included 2) Fraction collector F9-C: Covered cassette format x-y fraction collector for safe collection of up to 576 fractions in deep-well plates, tubes, and bottles. 3) UNICORN 6.3.2 WrkStn-Pure - BPL: UNICORN 6.3.2 Workstation (node-locked) e-license and DVD for AKTA pure, and AKTA pilot platform. One (1) license access code sticker on DVD case. Design of Experiment (DOE) and Column Logbook functions require separate e-license to activate Includes Microsoft SQL Express Edition (capable of supporting up to 3 client stations) Compatible with Windows 7 (64bit, 32bit) Professional and Windows XP Professional 4) Column Valve Kit (V9-C, 5 Columns): Switch automatically between 5 columns. Automate cleaning and increase eluted concentration using column bypass and reverse flow. Safeguard columns through pre-and post-column pressure measurements. 5) Superloop 10 ML AKTA: Superloop allows the introduction of large sample volumes into a pressurized fluid system. It is used together with the injection valve MV-7, INV-907, V9-Inj, or V9H-Inj and replaces a simple sample loop. 6) Superloop 50 ML AKTA: Superloop allows the introduction of large sample volumes into a pressurized fluid system. It is used together with the injection valve MV-7 or INV-907 and replaces a simple sample loop. 7) Superloop 150 Ml 8) HP8300 Elite SFF WIN7- 64Bit The Salient Characteristics of the AKTA Pure M1 Chromatography System is as follows: The following Salient Characteristics must be provided: • The system shall have low pressure protein purification system to purify proteins and other biomolecules • The system shall deliver a smooth, low pulsation flow to the column in increments of 0.1 ml/min up to a maximum of 25 ml/min at 0 to 20 MPa pressure. • The system shall allow users to perform time-based, volume-based and column- based programming. • The system shall have multi-wavelength UV/V's monitor and detects up to 3 wavelengths from 190 to 700 nm. This can purify, identify and distinguish the labeled protein from the unlabeled species since they absorb at different wavelengths. • The system shall have modular design, which brings versatility and flexibility in using the system. Additional parts/modules can later be added on to further expand the system capability and productivity. Due to the accessibility and design of the modules, easily changed, which allows quick and efficient customization. • The system shall allow for safe collection of 576 fractions in deep-well plates, tubes, and bottles. • The system shall contain a Column valve kit, which includes the V9-C column valve and 5-columns. The valve can control the flow of the column and monitor by measuring the change of pressure pre and post column. This is important since the column valve can adjust the flow rate according to the change in pressure, instead of the system abruptly stopping the flow due to pressure exceeding the upper limit. That means the run will continue as long as the flow rate is adjusted to keep the pressure under limit. The column valve kit also allows reverse flow through the column for fast and effective elution of strongly bound proteins, sharper peaks (increase purity), and a concentrated target molecule eluent. This is important for maximizing the yield for purifying the target protein for subsequent characterizations. A reversed flow also help lengthens the lifetime and improve the performance of columns by rinsing away debris on the top. In addition, the inlet selection of the system comprises 4 inlet positions in a single valve giving a convenient solution for automation of buffers. Also it can switch automatically between 5 columns and automate cleaning. • The system shall contain a Superloop 10 ml, 50 ml, and 150 ml AKTAs allow for the introductions of large sample volumes into a pressurized fluid system. II. Delivery: Contractor(s) shall deliver the item within 90 days after award to: NIH/NCI Bethesda, Maryland 20892. The Contractor that receives the award will be provided with the complete mailing address and point of contact. Upon delivery, contractor must notify the NCI Contracting Officer's Representative (COR) to schedule the installation date and time. III. Installation: To be carried out by the manufacturer and shall occur within approximately one week of delivery. IV. Payment: Payment shall be made after delivery, installation, and successful operation of equipment. Payment authorization requires submission and approval of invoice to the NCI COR and NIH Commercial Accounts. QUOTATIONS ARE DUE: September 10, 2013 at 11:00 am EST.
