- The Department of Commerce (DOC), National Institute of Standards and Technology (NIST), the Multiplexed Biomolecular Science Group (MBSG), conducts research in all aspects of measurement science, technology development, and standards for measurements where biological state is related to the contemporaneous levels of many biomolecules; establishes methods for asserting traceability, assessing measurement uncertainty, and validating measurements for multiplexed measurands; establishes approaches for measurement assurance of multiplexed and multivariate measurands, including standards and methods development; participates and leads relevant standards organizations and international bodies to develop and promulgate standards for and approaches to multiplexed and multivariate measurement assurance; develops multiplexed metrology for all facets of biochemical science, including clinical, pharmaceutical, biological and biomedical research, forensic, and homeland security applications. The MBSG is starting the development of new whole human and bacterial genomic reference materials for DNA sequencing and in collaboration with the Genome In A Bottle Consortium (GIAB). To meet its mission with confidence and to meet deadlines, the MBSG needs to procure an Ultra-High Throughput DNA Sequencing System. Throughput (Gigabases (Gbs)/run) is important because samples need to be run together for comparison purposes for certification measurements, e.g., homogeneity and stability of already obtained samples and future samples. The MBSG is conducting inter-laboratory measurements of genomic reference materials. For comparability between NIST measurements and measurements from other laboratories using the Illumina Hi-Seq 2500, it is necessary that NIST personnel can make measurements on an Illumina Hi-Seq 2500 in NIST laboratories. NIST,Acquisition Management Division (AMD) has a requirement for an Illlumina Hi-Seq 2500 w/cBot system. This requirement will be sole sourced to the following company: Illumina, Inc. The Illumina Hi-Seq 2500 w/cBot system shall have the minimum capabilities: 1. Capable of Paired End and Mate Pair sequencing 2. Capable is capable of sequencing both RNA and DNA libraries and both can be run simultaneously on a given run in Standard Mode 3. Library Preparation Requirements: a) Amplification-free library preparation option for DNA b) Mate Pair Library preparation with up to 15kb insert capability c) DNA and RNA library capabilities d) Provide a low-input (<10ng) DNA library preparation protocol 4. Capable of achieving the following read length: a) 2 x 150 bps in Rapid Mode b) 2 x 100 bps in Standard Mode 5. Capable of producing the following throughput (Gbs/run): a) 160 Gbs in Rapid Mode in ~2 days b) 600 Gbs in Standard Mode in ~ 11 days 6. Capable of producing 900 Gbs of mappable data in 30 days 7. Percentage of bases with Quality Values (Q) of 30 (inferred base call accuracy of 99.9%) or higher: a) Rapid Mode i. 2 x50 reads, ~ 85% ii. 2x100 reads, ~80% iii. 2x150 reads, ~75% b) High Throughput Mode i. 2x50 reads, ~85% ii. 2x100 reads, ~80% 8. Automated clonal cluster generation 9. Prepare 1 flowcell for HiSeq 2500 High Throughput run mode in 5 hours 10. Capable of preparing up to 96 samples per run 11. Requires <1 hour hands on time 12. Dell enterprise class 64-Bit machine 13. Windows 7 OS 14. Hardware must meet DOC ITSPP media sanitization requirements The NAICS code is: 334516. It is anticipated that one (1) Firm-Fixed Price, purchase order will be awarded for one year to include service warranty of instrument. This is not a Request for Quote. Sources interested in this requirement must provide a statement of capabilities in sufficient detail to determine if the requirements of this synopsis can be met. Responses must be in writing and must be received within seven (7) calendar days from the date of this notice. A determination by the Government not to compete the proposed purchase order based on responses from this notice is soley within the discretion of the Government. Information received will normally be considered soley for the purpose of determining whether to conduct a competitive procurement.
