Description
The NRL requests information on three detector analytical ultracentrifuge that have comparable or better characteristics than the specifications below: (a) High intensity xenon light source to provide continuous spectrum UV-visible light; (b) Optical components with an evacuated light path to reduce scatter and provide useful light for detection down to 190 nm so that proteins not readily detectible by other UV systems (static spectroscopy or HPLC) can be detected; (c) Wavelength range from 190nm to 800nm; (d) Photometric display range from 0 to 3 absorbance units; (e) A monochromator that features toroidally-curved holographic diffraction grating to provide monochromatic light over a wavelength range of 190nm to 800nm; (f) It is desired that the system have a minimum number of optical surfaces in the light path to minimize incidental absorption for increased sensitivity and accuracy; (g) The light source must have a flash lamp that illuminates the samples only during scanning. (h) The detector must provide automatic baseline correction and scanning slit/lens assembly that moves beneath the light to detect minute differences in molecular vs. radial distance; (i) The aperture setting must be selectable to either 2 or 4 degrees to provide precise detection at narrowly defined segments of the sample; (j) The interface optical system must contain its own separate light source, optical track and detector. (k) The light source must consist of a 30-m Watt laser diode operating at 675nm and must be directly attached to the monochromator; (l) The two slits must be located at the base of the laser so that light passes through the cell containing the reference and the sample and is picked up by the optical track located below the refrigeration can; (m) The detector must consist of a high resolution CCD camera with a resolution of 96 X 2048 pixels that permits a scan and a plot to occur in five seconds; (n) The interference optics and UV/Visible optics must be capable of working independently of each other or simultaneously; (o) The system must provide acceleration of 5 to 400 rpm per second with a maximum of 400 rpm acceleration; (p) The deceleration must be 5 to 400rpm per second with a maximum of 400 rpm deceleration; (q) The system must have temperature stability within 0.3°C of a set temperature with a control to 0.5°C and display to 0.1°C; (r) The system must have aluminum cells capable of handling samples in non-aqueous solvents; (s) The fluorescent detection system must meet the following minimum requirements: a. Be completely compatible with the main analytical centrifuge system; b. 488nm laser excitation and an emission range from 505 to 585nm; c. A lower detection limit in the 10's of picomolar range; d. A usable detection range spanning 4 decades above the lower detection limit for a fluorophore such as fluorescein; e. 1% signal linearity over the usable range; f. 50 micron radial resolution with data acquired at 2 micron intervals. (t) It is desired that the system have a minimum number of optical surfaces in the light path to minimize incidental absorption for increased sensitivity and accuracy.
