- THIS IS A NOTICE OF INTENT, NOT A REQUEST FOR PROPOSALS. A SOLICITATION DOCUMENT WILL NOT BE ISSUED AND PROPOSALS WILL NOT BE REQUESTED. The National Heart, Lung, and Blood Institute (NHLBI), Office of Acquisition (OA) on behalf of the National Center for Advancing Translational Sciences (NCATS), intends to negotiate and award a purchase order on a noncompetitive sole source basis to Lonza Walkersville, 8830 Biggs Ford Road, Walkersville, Maryland 21793. The items to be purchased include: 1. Part Number: AAU-1001, HT Nucleofector System, Quantity 1, 2. Part Number AWU-1001, HT Nucleofector Service, Quantity 1, 3. Part Number: AWT-1001 HT Nucleofector Installation and Training, Quantity 1. Sole Source Justification NACTS' RNAi facility provides intramural researchers with access to the equipment and expertise necessary to conduct genome-wide RNAi screens in mammalian cells. This facility works as a collaborative resource for doing RNAi assay development, screening, and initial follow up. The facility works with investigators to develop and optimize their assays for screening, conduct the screens and related informatics analyses, then perform follow-up screens on promising leads. Many of the facility's screening projects utilize lipid-based transfection. However, there are a number of projects that are refractory to lipid based transfection and require electroporation of siRNA. For example, any projects involving T cells or B cells require such methods. All of this work is currently performed using our Amaxa 96 well shuttle system (NIH decal 01812138), but is limited by the throughput of this system, thereby limiting our ability to conduct such projects. This request is for an Amaxa HT nuclecofector. This is necessary to improve the throughput of our screening operations, as it is 384 wells rather than 96. This format is also amenable to the format of our existing siRNA libraries, which will not only allow us to improve throughput, but also obviate the current step of having to re-array our libraries to 96 well format prior to using the existing shuttle system. Importantly, existing protocols optimized for the 96 well system can be directly translated to the 384 well system without re-optimization. Lonza, inc. is the sole company which develops and commercializes the Nucleofector® Technology, and to date the Nucleofector® Technology is not available from any distributor or dealer within the United States. Specifically, Nucleofector® Technology is a non-viral transfection method designed for primary cells and difficult-to-transfect cell lines. It is an integrated, turn-key methodology based upon three components: • The HT Nucleofector® Device and Nucleofector® 96-well Shuttle® System deliver defined electrical pulses to cells in suspension; • Cell-specific kits containing specially-formulated transfection solutions that condition Cells to facilitate transfection with the optimal pulse • Optimized protocols and experimental conditions detaining the handling of cells, pre, post, and during nucleofection® Lonza is the sole provider of the requested equipment. There is no other provider of a 384 well system that is amenable with NCATS' existing electroporation devices and corresponding protocols. This acquisition is being conducted under FAR Part 13, Simplified Acquisition Procedures, and is exempt from the requirements of FAR Part 6, Competition Requirements. The North American Industry Classification Code (NAICS) is 541711 and the Size Standard is 500 Employees. This notice of intent is not a request for competitive proposals. The determination by the Government not to compete the proposed purchase order based upon responses to this notice is solely within the discretion of the Government.
