- Description
- Description The National Cancer Institute (NCI), Cancer Center Research (CCR), Laboratory of Cell Biology (LCB) plans to procure the brand name Simon Simple Western System with Computer Compass software. The governments preference is to purchase refurbished equipment. This acquisition will be processed under FAR Part 12 - Acquisition for Commercial Items and will be made pursuant to the authority in FAR 13.106-1(b)(2) and 13.501-(a)(1) using simplified acquisition procedures for commercial acquisitions. The North American Industry Classification System code is 334516 and the business size standard is 500 employees. Only one award will be made as a result of this solicitation. This will be awarded as a firm fixed price type contract. Delivery Date: 90 days from date of award. NCI plans to purchase a fully automated complete solution for protein detection and characterization which performs all the manual processes associated with a traditional Western blot. Protein sizing, quantitation and immunodetection of proteins is performed in a nano¬immuno assay system that utilizes 12, 5 cm long, 100 pm inner diameter capillaries, with a ProteinSimple' patented inner coating that allows for the separation, capture and covalent immobilization of proteins from a wide array of species samples in a 40 nl volume, and subsequent immunoassay detection in the same vessel. The system is capable of analyzing 12 samples accessed from a microtiter plate with primary and secondary antibodies also accessed from this microtiter plate. The system uses fluorescent and biotinylated labeled molecular weight size standards and proprietary software to calibrate each capillary and assign molecular weight sizing to detected proteins. The immunoassay utilizes chemiluminescent detection and quantization of target proteins. The unique software allows the user to define the samples and assays to be assigned and then automatically performs sample uptake, separation, protein immobilization, immunoassay and chemiluminescent detection without further user involvement. Post run analysis software analyzes the chemiluminescent and biotinylated molecular weight standard data to assign molecular weight to target protein being immunodetected. Facilitates standardization of laboratory processes and provides data in a format that can be easily shared between multiple users and facilities. No other technology combines this unique method of physical separation and immunoassay detection in a nanoscale volume. Salient Characteristics • General Specifications • Bench-top footprint with dimensions not exceeding 46 cm H x 41 cm W x 74 cm D • Weight not to exceed 29.5 kg • Fully operable requiring only a single universal voltage (100VAC-230VAC, 50/60 Hz) power output connection • Fully operable in a humidity range of 15-70% relative, non-condensing • Fully operable in a temperature range of 16-270C • Uses a vacuum that requires 15L/minute at 15 inches Hg minimum • Processes a maximum of 12 samples per run • Run time 2-5 hours • Uses capillaries with a length of 5 cm • Uses capillaries with a volume capacity of 400 nL • Uses capillaries with a 100 inner diameter • Sample Input Specifications • Process a maximum of 12 samples per run • Store samples at room temperature • Analyze the following samples: culture mammalian cell lines, white blood cells, FACS¬sorted cells, plasma and crude cell culture supernatants • Reaction Vessel Specifications • Fused silica capillaries that are 5 cm in length, 100 tM inner diameter and 400 nL volume capacity • Perform protein capture using proprietary inner capillary chemistry coating that is UV light activated to cause covalent immobilization of all proteins to the inner capillary wall • Throughput Specifications • Process 12 individual samples in a ProteinSimple proprietary designed microtiter plate with space for separation and stacking matrices, primary and secondary antibodies and peroxide and luminol chemiluminescent substrate which are required to carry out 12 individual immunoassays Complete 12 nano-immunoassays within a cycle time in three to five hours Robotic automated systems which prepares capillaries for sample uptake • Robotic automated system which volumetrically loads 40 nL sample and - 360 nL of buffer • Robotic system which moves capillaries to electrophoresis station and orients capillaries to the anode and cathode reservoirs as well as to the detection station • Perform automated size-based separation Wash capillaries, remove matrices and perform automated immunoassay steps incubating of primary and secondary antibodies and protein blocking steps • Automatically introduce chemiluminescent reagents for immunoassay detection results • Separation Specifications • Molecular weight size separation power supply able to produce 0-3,000 volts • Constant voltage output • Immobilization Specifications • Software to control the UV lamp • Contains a low pressure mercury lamp • Detection Specifications • Automated detection of fluorescently labeled protein size standards • Sizing range of protein detection between 15-150 kDa • Ability to distinctly resolve biotinylated protein ladder standards • Chemiluminescent and fluorescent detector • CCD Camera that is cooled and with 1.5 megapixel and 16-bit resolution • Software Specifications • Compass software to set-up assay protocols and analyze run data • Specify up to 12 samples per run • Specify primary and secondary antibodies per sample • Specify the molecular weight per sample • Specify chemiluminescence collection time • Assign molecular weight values to each protein detected • Perform relative quantitation of identified peaks by total area or peak height • Export data to spreadsheet or other optional data analysis and presentation packages The data that was produced was previously unattainable using traditional Western methods due to the large sample size requirement and the non-quantitative nature of the traditional Western blot method. This is the first truly quantitative Western system on the market that can accurately quantitate endogenous levels of proteins in small volumes samples such as clinical specimens. The system is automated and greatly expedites the sample analysis time. Results are obtained in -2hrs with Simon vs 1 to 2 days with other Western methods currently available on the market. ProteinSimple is the patent holder of this technology and is the sole source manufacturer/distributer of this system. Potential offerors may request a copy of the solicitation from Erin Breedlove, Contract Specialist via electronic mail at breeerin@mail.nih.gov or fax (301) 402-4513 by August, 2012 11:00 am. Interested parties shall include the name of company; complete mailing address; point of contacts, phone, fax numbers, and email address; company DUNS number and business size. All questions shall be in writing and may be addressed to the aforementioned individual noted above. No collect calls will be accepted. In order to receive an award, contractors must be registered and have valid certification in the Central Contractor Registration (CCR) and the Online Representations and Certifications Applications (ORCA).
