Description
THIS IS A NOTICE OF INTENT, NOT A REQUEST FOR PROPOSALS. A SOLICITATION DOCUMENT WILL NOT BE ISSUED AND PROPOSALS WILL NOT BE REQUESTED. The National Heart, Lung, and Blood Institute (NHLBI), Office of Acquisition (OA) on behalf of the National Center for Advancing Translational Sciences (NCATS) intends to negotiate and award a purchase order on a noncompetitive sole source basis to Life Technologies, 2130 Woodward Street, Austin, Texas, 78744 for the purchase of one (1) Silencer Select Human Genome siRNA Library, V4, 384 -Well and one (1) Silencer Select Human Kinase siRNA Library V4, 384-Well. Background: The National Institutes of Health has established an RNAi facility to provide intramural researchers with access to the equipment and expertise necessary to conduct genome-wide RNAi screens in mammalian cells. This facility works as a collaborative resource for doing RNAi assay development, screening, and initial follow up. Our facility works with investigators to develop and optimize their assays for screening, conduct the screens and related informatics analyses, then perform follow-up screens on promising leads. Ambion Silencer Select siRNAs are used for the gene specific knockdown of mRNA transcripts in cells. This library has 3 independent siRNAs per gene arrayed in individual wells of 384 well plates. This item is needed to facilitate our RNAi screening program. This program conducts genome-wide siRNA screens in collaboration with NIH intramural investigators. This work includes RNAi assay development, screening, and initial follow up. The Ambion siRNA library will be used for genome-wide siRNA screens in human cells. Sole Source Justification: The standard offering of this library is a "pooled" format, which is not in line with NCATS general, more robust screening strategy of using one siRNA per well. Furthermore, NCATS has already optimized protocols for a number of our on-going studies using Ambion Silencer Select siRNAs and they have proven to be reliable reagents. NCATS will easily be able to extend these optimized conditions to genome-wide screening without expensive and laborious re-optimization. NCATS has also generated significant amount of data with the Ambion Silencer Select siRNAs which will be readily integrated with data generated from this genome-wide library. These requested siRNAs are the next generation Ambion siRNAs. These siRNAs incorporate the latest improvements in siRNA design, off-target effect prediction algorithms, and chemical modifications to yield siRNAs with unmatched efficacy, potency, and specificity. In siRNA screening applications, the result obtained is fewer false negatives due to poor silencing and fewer false positives due to off-target effects. In other words, with these siRNAs NCATS will get cleaner, more consistent data from our primary screen, so NCATS will expend less time and resources to find true, validated targets. Because of their importance in cell signaling and many biological pathways, kinases are key drug targets and subjects of intense scrutiny. The included Human Kinase siRNA Library V4 targets 710 human kinases with three individual per gene. This siRNA library enables systematic, yet cost effective, RNAi studies of these key cell regulators. Validated siRNAs are included where available. The included Human Kinase siRNA Library V4 is provided also provided in 384 well plates and will be employed in pilot screens prior to large-scale genome-wide efforts. These requested libraries contain 0.25 nmol of each siRNA, sufficient for 500 transfections (at 5 nM siRNA, 100 µl transfection volume). The siRNAs will be supplied dried into the wells of 384-well plates. The libraries are delivered with full siRNA sequence information, as well as the exon targeted and each targeted gene's symbol, full name, aliases, RefSeq number, and Entrez Gene ID, plus results from any validation experiments done using the included siRNAs. The North American Industry Classification Code (NAICS) is 325414 and the Size Standard is 500 Employees. This notice of intent is not a request for competitive proposals. The determination by the Government not to compete the proposed purchase order based upon responses to this notice is solely within the discretion of the Government.
