- Description/Specifications/Statement of Work Analyze protein expression changes in four different cell types (2 lung, 1 endothelial, and 1 heart) using aptamers as protein probes. Samples shall be analyzed so that the National Center for Toxicological Research can compare the effect of tobacco smoke condensates on the cellular expression of the proteins. Ultimately, the cellular expression changes of the various proteins will be used as biomarkers of exposure and harm to tobacco products. -- It is essential that the proteomics analysis uses aptamer technology and not another detection technology to provide a required high throughput, reliable, and quantitative technology. -- Shall use aptamers to detect 2:1000 proteins per sample 1) Samples will be shipped to the Contractor in 3 lots: 1 Lot of 40 samples will be shipped during the month of Sept 2012 1 Lot of 40 samples will be shipped during the month of Dec 2012 1 Lot of 30 samples will be shipped during the month of March 2013 2) The Contractor shall have 4 months from receipt of sample lot to run the samples and provide analytical data (protein expression levels) from the time they receive each lot. Data shall be sent at Vendor's expense to NCTR electronically and on a portable hard drive supplied by the Contractor. 3) Within 30 days after the conclusion of analyses any samples not utilized or leftover shall be packed on dry ice and returned at the Contractor's expense to NCTR. All samples may be batched into one shipment. Backgrougd Information Aptamers are single-stranded nucleic acids (ssRNA or ssDNA) that have the ability to bind a specific target molecule with high specificity and up to nanomolar affinities due to their 3D shapes.
