This announcement constitutes the only solicitation; proposals are being requested and a written solicitation will not be issued. We provide immediate and accurate remains identification in support of Operations Enduring Freedom (OEF) and present global conflicts. Our Research and Lab Bio-Automation missions, which include high throughput DNA analysis, have substantially increased our workload. Additionally, the AFDIL, an American Society of Crime Laboratory Directors (ASCLD) accredited forensic lab, provides mission essential services for a number of agencies, including the FBI, National Institute of Standards and Technology (NIST), the United States Army Criminal Investigations Laboratory (USACIL), and the University of North Texas Center for Human Identification (UNTCHI). The AFDIL collaborates with these institutions and is required to share data between these laboratories. The required laboratory equipment will enable AFDIL can continue the performance of their mission. The Armed Forces DNA Identification Laboratory (AFDIL) requires highly specialized, automated/robotic, liquid handling systems for post amplification processing. One instrument will be utilized by two different casework sections, to perform necessary post-amplification pipetting steps required for both mitochondrial DNA (mtDNA) control region sequencing and nuclear STR typing of reference samples. The second instrument will enable the automation of these tasks in addition to research projects such as entire mtDNA genome sequencing. The automation of these post-amplification steps is necessary to ensure efficient and accurate sample processing while meeting throughput requirements. To increase efficiency and enable the mtDNA Section to meet increased yearly sample processing goals, an effort to automate all case sample post-amplification laboratory processing and integrate this processing with the in-house LIMS system has been initiated. These improvements will decrease the amount of time required to process a single case sample as well as eliminate opportunities for manual errors. The automation and LIMS integration of these processes will increase throughput while decreasing the extensive user sample manipulation currently required, thereby allowing scientists to perform additional duties. The selected robotic instrumentation must meet both functional and software requirements for automation of these complex processes and must be suitable for communication with our in-house LIMS system. To further increase the capabilities of the instrument and decrease user interaction, the instrument must also have the potential to integrate cold storage of tubes and plates. Additionally, in preparation of new projects evaluating various Next Generation Sequencing (NGS) systems, like Roche's 454 GS Junior and Pacific Biosciences PacBio RS, the functions of the systems must accommodate preparation of indexed sample libraries for multiplexed sample analysis. Such library preparation requires multiple steps that when performed manually are extremely time-consuming and labor-intensive. The automation of the library preparation steps would greatly increase throughput as well as lessen user interaction, thereby freeing scientist time which can instead be devoted to data analysis. Limited physical space requirements for the additional peripherals and the ability to develop complex automated processes are imperative for the robotic instruments. SYSTEM CHARACTERISTICS Air displacement pipetting No dilution of samples with system fluid, no system liquid, diluters, valves or complicated tubing which can introduce bubbles that cause pipetting errors. Frictionless tip attachment Prevention of aerosol production during ejection, which is a potential source of contamination. No deformation to disposable tips therefore they can be reused an infinite number of times. Precise deck positioning Precisely and accurately reach any position on the deck without the need to "teach" the instrument and no need to redefine positions as coordinates drift over time. Independent movement on the Y and Z axes for the independent pipetting channels Greater flexibility for hit-picking of samples across multiple plates and/or tubes. Pipetting range (1-1000uL) for independent channels and 96-probe head Range required for the variety of assays to be implemented on the instrument. Certified accuracy for all pipetting channels, including the 96-probe head, down to 1uL. Liquid level detection for independent channels and 96-probe head Confirm the liquid level prior to or after pipetting in order to be able to troubleshoot downstream failure quickly. Training and method development support Users will be trained in the system software and method development in order to ensure their ability to use the software as necessary. Availability of advanced training Expert users have the option to take additional training courses to increase in-house knowledge and eliminate the need to utilize outside support in the future.
