Description
The Agricultural Research Service, on behalf of the Genomics and Bioinformatics Research Unit (GBRU), Stoneville MS, intends to negotiate and award on a non-competitive basis a purchase order to Covaris, Inc, 14 Gill Street, Unit H, Woburn, Massachusetts, 01801-1721 for the purchase of a S220 High Performance Ultrasonicator, pursuant to the authority of 41 U.S.C. 253(c)(1) and FAR 6.302-1, Only One Responsible Source and No Other Supplies or Services Will Satisfy agency Requirements. GBRU supports research projects involving DNA analysis of a wide range of species important to agriculture. In its operations GBRU is required to shear DNA for next generation sequencing, specifically the platforms HiSeq-2000 and Roche FLX. Prevention of cross contamination or to open air contamination are paramount to GBRU operations in the processing the samples. Thus closed containers for the shearing process are required and shearing needs to occur without the insertion of a probe into the DNA sample. Acoustic shearing of DNA using the Covaris S220 instrument produces fragments in a highly reproducible size (without major calibration efforts) range down to 100 bp; the fragment ends are easily blunt ended in a single enzymatic step; and sample volumes can be as low as 100 ul and sample mass as low as 100 ng. The process is isothermal, reducing bias to A-T region shearing. Required system operation specifications are as follows: 1. Computer controlled time and acoustic energy transfer and real-time monitored temperature. 2. The system frequency must be within the ultrasonic range (wavelengths on average less then 1mm in length), and thus outside the human audible range. 3. User-tuneable, generated fragment size distribution (range: 150-bp to 5-kbp) 4. Isothermal processing- no heat generation in the sample during processing.. 5. Closed tube, non contact process producing a controlled focused acoustic field inside a closed vessel. This is essential to limit sample cross-contamination and attendant contamination by other sources. 6. Sample input flexibility: tubes, vials, for single sample processing; and sample shearing, solubilization/disruption or extraction applications. 7. The maximum system size cannot exceed 21 cm wide, by 52 cm deep owing to space limitations. 8. System does not require hearing protection or segregation from the common laboratory environment. 9. The system can be integrated into liquid handling platforms for automated operations. 10. Can be used with an array of samples states such as liquid, solid, tissue, powder, soil, The settings and method profiles for this instrument are unique to the brand and model and this specific instrument is required to repeat experiments performed at USDA-ARS. The Covaris S220 instruments are the only commercially available instruments that form ultrasonic waves for DNA shearing and cell membrane destruction without requiring a probe. The settings and methods used on this instrument are unique to this specific brand and model. The S220 instrument is proprietary to Covaris and is not available through another known source. The Government intends to award a firm fixed price Purchase Order on or about August 29, 2011. The acquisition is being conducted under simplified acquisition using FAR Part 12 procedures and exempt from the requirement of FAR Part 6, Competition Requirement. This Notice of Intent is not considered a Request for Quotation (RFQ) nor is it a Request for Proposals (RFP). No solicitation document is available and telephone requests will not be honored. Interested persons may identify their interest and capability to respond to this requirement. A determination by the Government not to compete this proposed contract based on responses to this notice is solely within the discretion of the Government. The Government will consider all responses received by 4:00PM central time,August 26, 2011. Information received will be considered solely for determining whether to conduct a competitive procurement.
