- This notice is a combined synopsis/solicitation. The Government has a research based need for commercial items prepared in accordance with the format in Subpart 12.6, in conjunction with FAR 13.5, as applicable, and as supplemented with additional information included in this notice. This announcement constitutes the only solicitation; quotes are being requested and a written solicitation will not be issued. Submit offers on RML-RFQ-11012. The solicitation documents and incorporated provisions and clauses are those in effect through Federal Acquisition Circular 2005-53 dated 7/5/2011. This acquisition will be processed under Simplified Acquisition Procedures (SAP) and is a Total Small Business Set-Aside. The North American Industry Classification System (NAICS) code for this procurement is 325414 and the small business size is 500. SCHEDULE: The National Institute of Allergy and Infectious Diseases (NIAID), Rocky Mountain Laboratories (RML), performs basic research contributing to the development of diagnostic reagents, therapeutics and vaccines against human pathogens. The RML has a research need for two recombinant Ixodes scapularis tick proteins to be generated so that vitro and in vivo studies will determine if they affect flavivirus infection in cells or in an animal model. These tick proteins appear to be glycosylated, so they must be generated in a system that will add the appropriate post-translational modifications onto the proteins. As such, we require these proteins to be generated in an insect based system so they very closely resemble what they would look like if they were produced in ticks. The two protein sequences must be codon optimized, synthesized and cloned into a Baculovirus shuttle vector. Production reports must be submitted after these steps. These sequences must be transfected into insect cells to generate a baculovirus stock for each protein. A Multiplicity of Infection timecourse must be performed to determine the optimal time to harvest the virus. Cultures of insect cells must be maintained to generate recombinant virus expressing our protein of interest. Finally, the proteins of interest must be purified by affinity chromatography, have the affinity tag cleaved off by enzymatic methods and the final protein polished to remove contaminants so it will be suitable for use in our experiments. The final deliverable for this order to be the entire yield of purified protein - approximately 5L equivalent.FOB Point shall be Destination, Hamilton, MT 59840. Delivery location is Rocky Mountain Laboratories, 903 S. 4th Street, Hamilton, MT.
