- PCR System Doc Work Statement for the Purchase of a OneStepPlus Real-Time PCR System for Molecular Microbial Source Tracking Multi-Lab Methods Validation at NOAA/AOML. Item: Real-Time Quantitative 4-color Rapid PCR Thermocycler system (ABI OneStepPlus) for use in the AOML Environmental Microbiology Program Multi-Lab Methods Development and Validation in Environmental Molecular Microbial Source Tracking. Category: Oceanographic and Marine Biology Scientific Equipment Number of units: 1 Narrative: As part of the Environmental Microbiology Program collaborative methods and biosensor development and validation research in environmental molecular microbial source tracking, AOML is involved in a variety both on-going and pending methods comparison and validation studies with a variety of Federal, State, and local agencies, and academic research institutions. As part of these studies, AOML needs to provide molecular microbial source tracking data for a variety of quantitative PCR assays conducted in an identical fashion to the other national labs involved in these studies. By group consensus, all labs involved in these studies MUST use exactly the sample qPCR thermocycler instrument, the same reagents, and the same methods protocols in conducting these comparative assays to ensure uniformity and validity of data for assessment and comparison of methods' precision, reproducibility, and effectiveness between different operators and locations. The instrument that has been selected by consensus with US EPA and the study labs, so as to provided consistency of instrumentation across all labs participating in these methods comparison studies Technical Criteria: The specific technical criteria designating this particular quantitative PCR instrument make and model was determined by the consensus of the US EPA and the collaborative participating labs in these comparative methods studies to eliminate variables of different instrumentation platforms, reagents, or protocols during the inter-lab comparisons. Overall System Description: The real-time quantitative PCR thermocycler system to be purchased must include the following parts and components with the system package: (1) OneStep Plus 96-well real-time quantitative PCR thermocycler, ABI part number 4376598. (2) Laptop computer, equivalent to a Dell Laptop D530, set up and configured to control and acquire data from the OneStep Plus PCR instrument, and including the following software listed below pre-installed. (3) All connection cabling and peripherals necessary for data acquisition by the laptop from the OneStep Plus PCR instrument. (4) StepOne software v2.1 or greater pre-installed on laptop. (5) Primer Express Software v3.0 or greater pre-installed on laptop. (6) OneStep Plus packing kit. (7) All user manuals for OneStep Plus PCR instrument, laptop, and software. (8) FG, FAST 96-well RNase P Verification plate, part number 4351979. (9) Total of 4 years of warranty on parts and labor StepOnePlus, part number ZGEXSCSTEPONEP.
