- Justification & Approval Program Office: NHGRI Title: Period of Performance: July 30, 2010- December 31, 2010 Proposed Contractor: Sigma Aldrich Inc. Description of what is to be acquired and the Benefit received by the Government: NHGRI Zebrafish Core facility identifies mutations in genes involved in human genetic diseases for functional studies by NHGRI investigators. To complement our on-going resequencing project, we are using recently developed targeted mutagenesis strategy using zinc-finger nucleases designed specifically for the target gene. Main Objective of generating knockout mutants is to generate disease models that can help in understanding of gene function, disease pathology and development of therapeutic approaches. In this case, genes involved in human genetic diseases are studied in an animal model (zebrafish) for better understanding of the disease mechanism. For this project, Sigma-Aldrich (Vendor) will design and test several ZFN pairs for two target genes identified by the customer (Zebrafish Core). They will use Sangamo SuperFinder algorithm to design specific ZFN candidates, assemble them using ZFP modules from the proprietary Sangamo ZFN archive, such that a target-binding specificity of at least 24 bp is achieved. They will attach a DNA-cleaving Nuclease domain to the assembled ZFP and then validate them by mismatch sensitive nuclease assay. The ZFN pair with the highest DNA-cleavage efficiency will be provided to the Core (Government) as ZFN plasmids and 10 Aliquotes of Ready-to-Deliver mRNA. This project will begin in July 2010 and estimated to be completed by October 2010. Specific, detailed explanation of why it is not feasible to obtain full and open competition. Sole Source Justification. Sigma-Aldrich is the only vendor that currently holds exclusive commercial rights to generate zinc-finger nucleases to target specific genes in Zebrafish. The technology was initially developed by Sangamo Biosciences and has been licensed to Sigma-Aldrich for sale to research community. NHGRI Zebrafish Core provides services to NHGRI researchers that facilitate analysis of genes involved in human genetic diseases using zebrafish as a model system. To this effect, we have been performing targeted re-sequencing of ENU-mutagenized males (TILLING) for identification of mutations. This technology is limited by the number of males screened and the size of the exons as ENU generates mutations throughout the genome randomly. Therefore, for genes where TILLING has not identified knockout mutations, zinc-finger nuclease technology can be used for specific targeting. Therefore, Zebrafish Core is ordering zinc-finger nucleases for 2 genes selected from 4 proposals by the faculty. Statutory Authority. Federal Acquisition Regulation (FAR) Subpart 13.106-1. (b) Soliciting from a single source. (1) For purchases not exceeding the simplified acquisition threshold, contracting officers may solicit from one source if the contracting officer determines that the circumstances of the contract action deem only one source reasonably available (e.g., urgency, exclusive licensing agreements, or industrial mobilization). No other requirement and/or type for supply or service will satisfy agency need. Market research: The review of previous purchase orders for the identical requirement.
