- The U.S. Department of Commerce (DOC), National Oceanic and Atmospheric Administration (NOAA), National Marine Fisheries Service, located in Miami, FL intends to acquire analytical services to discern the degree of genetic variation in temperature tolerance of planktonic larvae of Endangered Species Act (ESA)-listed elkhorn (Acropora spp). Period of performance is (12) twelve months. The contractor shall be conducting experiments in Aug-Sept 2010 with larvae collected/fertilized from Acropora parents of known genotypes in Key Largo, FL, exposing them to three different temperature treatments throughout their pelagic larval duration. Under this contract, the contractor will analyze, and assist in interpretation of results, the temperature-stress gene expression of these (Florida) larvae to discern if Florida larvae have similar response to temperature as Puerto Rico larvae. Results will help elucidate the amount of genetic variation in this imperiled coral which could potentially provide scope for adaptation to global climate change. Requirements: There are two tasks under the contract TASK 1: Provide multilocus genotypes for up to 500 coral colonies (potential parents for the larval experiments and other members of the population) This contract requires analytical services to determine the multilocus genotype of up to 100 Acropora palmata tissue samples (ethanol-preserved) provided by SEFSC according to the methods of Baums et al. (2005). Results will be provided to SEFSC in a spreadsheet. TASK 2: Microarray analysis and interpretation The contractor shall analyze larval gene expression of A.palmata larvae from 2010 Key Largo temperature exposure experiments utilizing the novel microarray developed from temperature exposures of A.palmata larvae in Puerto Rico (2008-2009). Results will be provided in a written report. The government knows of only one source Dr. Iliana Baums at the Pennsylvania State University. Dr. Iliana Baums is the developer o the microsatellite genetic marker system which has been validated and documented to distinguish genotypes of A.palmata with high confidence. Dr. Baum's has published results on this method and the only known source that can reasonably be expected to complete the work efficiently and accurately. The A.palmata larval microarray is the completely novel took that Dr. Baums has under development. The specifications of this method have not yet been published and Dr. Baums' is the only known laboratory that can apply this genomic tool to elucidate the genomic response of ESA listed corals in the Florida temperature exposure experiments. To be considered, firms must furnish detailed information concerning their education, experience and capability to provide the services and satisfy or exceed the Government's needs. Such information shall include, at a minimum, name, telephone number and address of concerns for whom you have furnished this type of services previously, and detailed narrative that described your facility and ability to provide this service. Interested parties must respond within seven (7) days after publication of this notice. The government invites firms that believe they have the capability to meet the requirement to response, providing specific information that will enable the government to determine whether or not to compete the requirement.
