Description

The USDA, ARS, Knipling-Bushland U.S. Livestick Insects Research Laboratory, Kerrville, Texas intends to solicit and negotiate with only one source under the authority of FAR 6.302-1(a)(2)(ii)(A) to Amplicon Express, 2345 NE Hopkins Court, Pullman, WA 99163. This procurement is being conducted under Simplified Acquisition Procedures (SAP) - a follow-on contract for the continued development or production of a major system or highly specialized equipment, including major components. Amplicon Express synthesized our current BAC library (which consists of 47,000 clones and 5-384 well plates. Amplicon Express also has in their possession from the original synthesis, the prepared genomic DNA from the Deutsch strain of Boophilus microplus ticks that is necessary for synthesizing the library. The tick material from this strain is in low supply so by using the prepared material which Amplicon Express has saved from the previous library, we save precious Deutsch tick strain material for other research need. The following is the minimum work required: We require the synthesis, plating, secure storage of, exclusive license for, and a working copy on 384 well plate formats and arrayed nylon filters of a BAC library from Boophilus microplus genomic DNA from the Deutch strain of Boophilus microplus. We require synthesis of a library containing 147,456 clones arrayed onto 384 well plates. As clones are picked into the 384 well plates, we require quality control checks and identifications of all clones that did not grow. We also require RE-GRlDS of all 384-well plates of picked clones onto agar XGALlIPTG plates and re-grows to calculate the percentage of BLUE CLONES (without inserts) in the library. We require guarantees the total 'error rate' in the library (missed wells + blue clones) will be below 3%. All QC data should be conveyed to us in accessible file format. We also require clones to be gridded onto nylon membranes to verify colony morphology is correct. We also require a working copy of the library on 384 well plates, the company maintain a copy of the library in their laboratory -80 deg C freezers (with C02 back-up, auto dialers, alarms and insurance) as a backup, and the company provides us with a set of 8 nylon filters as a copy of the library suitable for screening in our laboratory. We also require exclusive lifetime rights to the use of the BAC library. The Government intends to acquire a commercial item using FAR Part 12. Interested organizations may submit their capabilities and qualifications to perform the effort in writing to the identified point of contact above not later than 3:00pm CST on March 23, 2010. Such capabilities/qualifications will be evaluated solely of determining whether or not to conduct this procurement on a competitive basis. A determination by the Government not to compete this proposed effort on a full and open competition basis, based upon responses to this notice, is solely within the discretion of the government. Oral communications are not acceptable in response to this notice. No RFP will be issued based upon this Notice of Intent.
