Description
Material specification: This material is needed for large scale trials and evaluations of various dsRNA constructs previously designed for the purpose of mosquito population control. In the development of RNAi based products, it is normal that adjustments and “fine tuning” will be required to improve efficacy of the initial sequences. The material is a dual stranded Ribonucleic Acid (dsRNA) specific and predefined sequence of approximately 350 nucleotides long. The exact and complete sequence to be produced (template) will be provided by the USDA/ARS scientists in a standard molecular description. If it will be necessary to produce significantly longer sequences, the vendor may be needed to generate sequences double this size. This template will be used to produce a batch of several grams according to order which will be delivered in cooled buffer solution in concentration of at least 1mg/ml dsRNA Quality check that ensures the produced material is to specification will accompany every batch produced and will be a condition and a proof of delivery. It is understood that the producing a construct to specification does not guarantee efficacy and it is part of the development process which may need consequent batches to be produced in similar process with a new order. 2. Intended use of items with respect to work involved, project duration, period of performance, and necessity for including specific requirement or characteristics. Intended Use The materials will be use exclusively for the research and development of RNAi based mosquito control agent within the USDA/ARS laboratories and under the supervision of Dr. James Becnel. Initial batch is expected to be used in the last few months of 2009. Additional batches may be necessary to be produced before the completion of the project. This material has not been through the regulatory phase and is not for any commercial use. Any further development based on the material which intended for commercial use will need the vendors’ certification and active participation in the commercialization process. From the time of final construct structure sign-of, the production of several grams batch will take on average 60 days and will be delivered on dry ice in express mail services. Documentation that includes clear storage and handling specifications will accompany each and every batch delivery. ***WORK STATEMENTS CONSULTING @ BLOCK OF 10 HOURS: 1. What needs to be done? The following are the consulting services that the project leader can use the vendor’s personal for: Review and inspect construct development strategy and make recommendations Inspect proposed sequences’ design vs. silenced target and make recommendations Assist in construct design adjustment as a result of trial analysis Assist in trial protocol development and their execution Other RNAi research and development expertise as required 2. Where should it be done? All consulting services will be delivered by electronic communication and do not mandate physical presence in the USDA/ARS offices. However, in case that such presence is required, a special arrangement that include work hours calculations, travel and expenses will be addressed prior to such assignment. In such case, it is understood that all additional expenses will be fully covered by the USDA/ARS. 3. What should the final output consist of? As the consulting services will be directed and used on needed basis by the USDA/ARS staff, these services will be delivered in good faith and to the best of the vendor’s knowledge. The will deliver the sequence but does not guarantee that at the end of the project any product of any kind will be complete or prove to be effective. 4. How will the final output be inspected/accepted? The final product will be tested with protocols that will be developed as part of the entire project. dsRNA according to certificate of analysis.
