Description
The U.S. Department of Commerce (DOC), National Oceanic and Atmospheric Administration (NOAA), National Marine Fisheries, Protected resources Division, St. Petersburg, FL intends to award a Purchase Order to The University of Southern Mississippi, Hattieburg, MS to: 1)establish the degree of genetic structure among Gulf sturgeon populations 2) Characterize interdrainage movement patterns of adults by genetic assignment tests and compare these results with the available capture/recapture history of individuals,and 3). Estimate population genetic parameters (e.g. effective population size) and demographic history and interpret these results in light of current estimates of population size and history as available for each drainage. The University of Southern Mississippi houses the Gulf sturgeon tissue archive, which has been accumulating samples generously provided by agency and academic researchers for almost nine years. Dugo et al. (2004) used some of these samples in their study focusing on population structure within the Pascagoula and adjacent drainages, but the collection has more than doubled in size since then (Table 1); eight hundred and thirty-two (832) samples have yet to be genotyped from this archive. The current archive not only represents more samples than reported on by Dugo et al. (2004), but it also provides coverage of two drainages that lack any published genetic data (the Suwannee and Ochlockonee Rivers). Various Gulf sturgeon researchers, as part of their normal research protocol, routinely take a small piece of fin tissue from individuals captured as part of their on-going studies. These tissue samples (i.e. fin clips) are immediately placed into a preservation buffer (Seutin et al., 1991) for transport to the lab where they are then stored at -20C. Gulf sturgeon are normally captured either in the summer holding areas of their respective drainages or in lower portions of rivers during the inward or seaward migrations during the spring and fall respectively. In addition to information of collection location, for most samples we also have record of other data recorded in the field (e.g. tag numbers, weight, total length and fork length). The government anticipates receiving approximately 300 additional samples over the course of the first year of the project from the following drainages: Apalachicola, Suwannee and Pearl; Sulak and Randall (USGS): Choctawhatchee; D. Fox (DSU) and F. Parauka (USFWS). Tasks Include: 1. Project planning - At the beginning of the project (September 2009), contractor will review project tasks and brief all personnel on the scope of the project and their duties. 2. DNA extractions - DNA extractions on current holdings of tissue samples will be conducted throughout October through January. New samples from the governments various cooperators will be processed as they are made available. Once made a start on these extractions, this work will be completed in conjunction with the genotyping (task #3, below). 3. Genotyping -Most of the laboratory protocols have already been optimized progress can be made in genotyping individuals throughout the remainder of FY2009-2010. Some additional time will be required to optimize protocols for the four new loci that will be added. 4. Data analysis -Conduct a preliminary analysis of the data in time for the first annual report in August (see #5 below). 5. Report preparation - The first report will provide an update on the state of our sampling processing and a preliminary data analysis. 6. Sturgeon meeting - The contractor shall attend the annual Gulf sturgeon meeting to brief participants on this project and seek their assistance in acquiring additional tissue samples. To be considered, firms must furnish detailed information concerning their capability to provide the services and satisfy or exceed the Government's needs. Such information shall include, at a minimum, name, telephone number and address of concerns for whom you have furnished this service previously, and detailed narrative that described your facility and ability to provide this service. This notice is for a service for which the Government intends to solicit and negotiate with only one source The University of Southern Mississippi. The Eastern Region Administrative Division (ERAD), requires that all contractors doing business with the Acquisition Office must be registered with the Central Contractor Registry (CCR).
