Description
The Centers for Disease Control and Prevention intends to issue a sole source purchase order to Lonza Walkesville, Inc., 8830 Biggs Ford Road, Walkersville, MD 21793-8415 for Amaxa Nucleofector 96-Well Shuttle, which includes Laptop. Research developed within the Special Pathogens branch has advanced our understanding of molecular virology with the use of reverse genetics and RNA interference. Both technologies demand efficient delivery of nucleic acid into cultured cells with minimal impact on cell viability. In the case of reverse genetics the goal is to launch infection of a genetically recombinant virus to experimentally determine components of the virus genome in virus assembly, replication, and assembly. RNA interference technologies allow the researcher to investigate what and how cellular pathways are involved with virus infection. The Amaxa Nucleofector transfects nucleic acid in a 96-well format using optimized conditions to ensure the highest degree of nucleic acid delivery and cell viability, which is necessary for work within the Special Pathogens branch. Lonza, inc. is the sole company which develops and commercializes the Nucleofector Technology, which is a non-viral transfection method designed for primary cells and difficult-to-transfect cell lines. It is an integrated, turn-key methodology based upon three components: the Nucleofector Device (or System) that delivers defined electrical pulses to cells in suspension; cell-specific kits containing specially-formulated transfection solutions that condition cells to facilitate transfection with the optimal pulse; and optimized protocols and experimental conditions detaining the handling of cells, pre, post, and during nucleofection. Nucleofection has been used successfully to transfect over 400 different cell lines and 100 primary cells at levels of efficiency, viability, and functionality unachievable by traditional chemical and electroporation methods. It is particularly effective transfecting non-dividing cells because, unlike other methods, no cell division is required for stable integration. This non-viral methodology delivers substrate to the cytoplasm and the nucleus making it suitable for stable clone generation in addition to transient transfection. Nucleofection has delivered DNA, RNA, siRNA, shRNA, Morpholinos, peptides, proteins, antibodies and small molecules making it a versatile tool that can be used throughout project enabling maximum amount of information to be gained from any particular cell type. The technology can easily co-transfect substrates, and unlike other transfection methods, Nucleofection allows the detection of expression in as few as 3 hours. CDC believes that this requirement is met by only one provider. This procurement will be processed under the authority of FAR 6.302-1 and 6.302-2. Only one responsible source and no other sources will satisfy agency requirements. No solicitation is being issued.
