- The U.S. Environmental Protection Agency (EPA), Ecosystems Research Division (ERD), 960 College Station Road, Athens, GA 30605 intends to award a fixed-priced purchase order for a Real-Time Polymerase Chain Reaction (PCR) System. The intent of this solicitation is to procure one PCR System meeting the minimum technical requirements specified below: Purpose of Instrument: Nucleic acid sequence detection by real-time polymerase chain reaction analysis Specifications of Requested Instrument: ? The instrument must be able to perform both real-time PCR (polymerase chain reaction) and post-PCR (end-point) analysis. ? The instrument must be capable of analyzing 96 samples simultaneously in a 96-well plate format as well as individual tubes. ? The instrument must provide application specific software to collect and analyze fluorescence data to be able to perform absolute quantitation, relative quantitation, allelic discrimination/SNP (Single Nucleotide Polymorphism) detection and plus/minus assays utilizing internal positive controls. ? The instrument must support two homogeneous reaction chemistries, the fluorogenic 5? nuclease assay using TaqMan?? probes and the SYBR?? Green I double stranded DNA binding dye chemistry. ? The instrument must utilize a tungsten-halogen lamp, a cooled charge coupled device (CCD) camera, a heated lid assembly, a Peltier heating/cooling system, and a system that includes multiple filters sets to enable multiple wavelength detection. ? The instrument software should be able to simultaneously detect at least 4, optimally 5 fluorophores with reduced crosstalk to be able to perform multiplex quantification of at least 4 target sequences and an internal standard. ? The system must be provided with a dedicated tower computer system including flat monitor or offer optional notebook computer set-up to reduce laboratory footprint. ? The instrument must be capable of performing both <2 hr standard analysis time or <40 minute rapid Real-Time PCR assays and be capable of resolving 2-fold difference in target sequence copies with statistical significance. The instrument must be able to use fast master mix reactions for true rapid real time analysis. ? The system should be able to detect at least 10 target sequence copies, and it should have a dynamic range for target sequence quantification of at least 5 orders of magnitude, it must include assay design and optimization guidelines and be supplied with primer and probe design software. ? Installation of equipment, on-site training and repairs should be provided by a factory trained service engineer. The system must be provided with at least one year of warranty. The vendor must be able to offer Telephone Technical Support and Field Applications/Sales/Service Support to help solve chemistry and instrumentation problems encountered with real-time quantitative PCR. ? Section 508 Compliance data for hardware and software The NAICS code for this procurement is 339111, size standard: 500 employees. The acquisition is unrestricted in regards to business size. Anticipated award date is May 15, 2007. All items shall be delivered to US EPA ERD, 960 College Station Road, Athens, GA within 60 days after receipt of order, FOB Destination.
