Description
This is a combined synopsis / solicitation for services being procured in accordance with FAR Part 13, Simplified Acquisitions Procedures. NOAA, National Marine Fisheries Service (NMFS) located in Seattle, Washington intends to acquire the following services: Statement of Work . Background and Objective: The NWFSC provides scientific and technical support to NOAA Fisheries for the management, conservation, and wise use of the Northwest region?s marine and anadromous species. The Conservation Biology Division provides technical support and conducts research related to the conservation of marine organisms. The objective of this project is to determine if there are cryptic species within the single currently recognized northern abalone species (Haliotis kamtschatkana ), by assessing levels of variation in "speciation genes" which mediate species-specific fertilization in abalone. The northern abalone is considered a potential candidate for ESA listing due to a severe decline in abundance that has occurred over the last several decades. An essential first task in conducting an ESU status review is to correctly identify the unit that is being considered for listing. For invertebrates, such as abalone, this unit must be a species or subspecies. Description: Task1: Sample collection. Obtain as many samples of northern abalone as possible (and no fewer than 20), focusing on extremes of the range. Samples may be collected by the vendor and/or obtained from existing collections. Newly collected samples shall consist of small tentacle clips that do not harm the individual sampled. Task 2: Genetic analysis. DNA shall be purified using standard methods. The genes for egg VERL and its interacting sperm protein lysin have already been sequenced from northern abalone [4,5], and these sequences shall be used for designing species-specific primers. Additionally, abalone-specific mtCO1 primers have been developed [6], and these shall be used as well. The contractor shall PCR and sequence the N and C terminal regions of the VERL gene, the coding sequence of the lysine gene and the mtCO1 gene from all individuals collected in Task 1. Sequencing shall be be performed using standard methods (e.g., ABI BigDye v3.1 an analyzed on an ABI3100 automated sequencer). The contractor, in consultation with the NWFSC point of contact, shall test for population differentiation using standard phylogenetic and population genetic criteria (e.g., those described in the documentation to the computer program PHYLIP (http://evolution.genetics.washington.edu/phylip.html). Deliverables Monthly phone or email updates to the CBD point of contact on the status of tasks 1 and 2. 3.2 A final report in standard scientific form (methods, results, discussion), including an electronic database of all data collected. The database shall include processed sequence data from all genes collected for all samples, all sequencing electropherograms, and all data related to sample collection (exact location and date for each collected individual). All individuals sequenced shall be identified with a unique numeric code, and this code shall be associated with all data collected on the individual. . 2 Performance period. October 2008 Final report December 2008 6.0 Data rights Upon collection, the contractor shall have the right to analyze, interpret, and publish the data collected under this statement of work without further consultation with the NWFSC. Upon receipt of the final report, the NWFSC shall have the right to analyze, interpret, and summarize the data in unpublished reports, white papers, ESA status reviews, and similar unpublished forms of scientific writing (i.e., ?grey literature?). The NWFSC recognizes the intellectual contributions of the vendor to the data, and will refrain from publishing in the primary scientific literature any of the data or analysis based upon the data until the vendor has done so, or until three years after receipt of the final report, whichever comes first. Upon publication in a scientific journal or at a time three years after submission of the final report (which ever comes first), the vendor shall submit all data collected under this statement of work to the publicly available DNA sequence database base Genbank (http://www.ncbi.nih.gov/). This announcement constitutes the only solicitation, quotatios are being requested and a written solicitation will not be issued.
